Article Synopsis
  • The study analyzed high-grade intraepithelial lesions (LIEHG) from 2010 to 2018 in a large population of asymptomatic women aged 25-65 who participated in a cervical cancer screening program.
  • Out of 305,940 women, 3,230 were diagnosed with LIEHG, showing a prevalence of 0.4%, with detection rates significantly varying by age—particularly higher in women under 35.
  • The results suggest that follow-up for low-grade smears is crucial and that adherence to screening recommendations lowers the risk of LIEHG, especially among younger women.

  • VEXAS syndrome, identified in 2020, is caused by mutations in the UBA1 gene and shows a variety of clinical and hematological features, making it challenging to distinguish from other inflammatory conditions. !* -
  • This study collected a dataset of 9,514 images of polymorphonuclear cells (PMNs) and used a convolutional neural network (CNN) to automate the detection of specific dysplastic features unique to VEXAS, achieving a high level of accuracy (AUC of 0.85-0.97). !* -
  • Results indicate that automated analysis can effectively support hematologists in identifying potential VEXAS cases, suggesting a screening score for UBA1 mutational

Article Synopsis
  • The study examines outcomes of 67 patients (average age 20.6 years) who underwent their first allogeneic hematopoietic stem cell transplant (HSCT) due to GATA2 deficiency across 21 centers in June 2022, showing varied indications for the transplant.
  • The findings reveal significant rates of acute graft versus host disease (GvHD) and chronic GvHD; the incidence of relapses was low, with overall survival rates at 1 and 5 years being 83% and 72%, respectively.
  • The analysis highlights that monitoring bone marrow for clonal evolution is crucial to initiate HSCT before the development of excessive blasts, noting that factors like earlier years of HSCT and certain

Objectives: X-linked hypophosphatemia (XLH) is characterized by increased concentrations of circulating fibroblast growth factor 23 (FGF-23) resulting in phosphate wasting, hypophosphatemia, atypical growth plate and bone matrix mineralization. Epidemiologic studies suggest a relationship between FGF-23, obesity, and metabolic dysfunction. The prevalence of overweight and obesity is high in children with XLH.

Predator responses to warming can occur via phenotypic plasticity, evolutionary adaptation or a combination of both, changing their top-down effects on prey communities. However, we lack evidence of how warming-induced evolutionary changes in predators may influence natural food webs. Here, we ask whether wild fish subject to warming across multiple generations differ in their impacts on prey communities compared with their nearby conspecifics experiencing a natural thermal regime.

Article Synopsis
  • Graft-versus-host disease (GvHD) is a serious problem that can happen after getting a stem cell transplant.
  • Researchers found that a special kind of cells called DP8α Tregs, which help control the immune system, might help prevent GvHD by linking to helpful gut bacteria.
  • The study showed that patients with fewer DP8α Tregs were more likely to develop GvHD, suggesting that boosting these cells could be important for preventing this disease.

The impact of pre-graft COVID-19 vaccinations in donor or recipient as well as pre-graft infection has been studied in 157 adults having received allogeneic stem cell transplantation (Allo-SCT) for various hematological diseases during the delta/omicron waves. We showed here that pre-Allo-SCT COVID-19 vaccination and/or infection do not provide more protection in patients receiving vaccine, immunotherapy or both after transplant. COVID-19 vaccination is and remains of crucial importance after Allo-SCT, reinforcing the recommendation to start COVID-19 vaccination as soon as the third month following the transplant.

In the last 20 years, several techniques have been developed for quantifying DNA methylation, the most studied epigenetic marks in eukaryotes, including the gold standard method, whole-genome bisulfite sequencing (WGBS). WGBS quantifies genome-wide DNA methylation but has several inconveniences rendering it less suitable for population-scale epigenetic studies. The high cost of deep sequencing and the large amounts of data generated prompted us to seek an alternative approach.

In Canada, a majority of federal constituency offices deal primarily with immigration files. The few qualitative studies on the subject show that the resources dedicated to these files and the type of work carried out on the immigration files handled vary between offices, thus contributing to disparities in service between federal electoral districts. How can such variation be explained? Based on the quantitative analysis of unpublished administrative data, this article first highlights the diversity of files handled by constituency offices, as well as the types of intervention carried out by constituency assistants.

Because they hold information about cultural identity, foodways have been the focus of a variety of disciplines in archaeology. However, each approach documents different stages of culinary preparation and is constrained by the preservation specificities of each type of artefact and ecofact. Difficulties in achieving an interdisciplinary approach may explain the scarcity of such studies.

Objectives: Vacuoles, E1 enzyme, X-linked, autoinflammatory and somatic (VEXAS) syndrome is an adult-onset autoinflammatory disease associated with somatic ubiquitin-like modifier-activating enzyme 1 (UBA1) mutations. We aimed to evaluate the efficacy and safety of targeted therapies.

Methods: Multicentre retrospective study including patients with genetically proven VEXAS syndrome who had received at least one targeted therapy.

Currently, no effective vaccine to prevent human immunodeficiency virus (HIV) infection is available, and various platforms are being examined. The vesicular stomatitis virus (VSV) vaccine vehicle can induce robust humoral and cell-mediated immune responses, making it a suitable candidate for the development of an HIV vaccine. Here, we analyze the protective immunological impacts of recombinant VSV vaccine vectors that express chimeric HIV Envelope proteins (Env) in rhesus macaques.

Late relapse, beyond 2 years following alloHSCT for AML, is rare. Among the 376 patients allografted for AML in our center between 1990 and 2016, 142 (38%) relapsed. The majority (68%) of relapses occurred during the first year following transplantation.

Anthrax toxins are critical virulence factors of and strains that cause anthrax-like disease, composed of a common binding factor, the protective antigen (PA), and two enzymatic proteins, lethal factor (LF) and edema factor (EF). While PA is required for endocytosis and activity of EF and LF, several studies showed that these enzymatic factors disseminate within the body in the absence of PA after intranasal infection. In an effort to understand the impact of EF in the absence of PA, we used a fluorescent EF chimera to facilitate the study of endocytosis in different cell lines.

  • VEXAS is a newly identified syndrome linked to mutations in the UBA1 gene, which may lead to an increased risk of disseminated non-tuberculous mycobacterial infections (NTMi), despite not being officially recognized as a cause of acquired immunodeficiency.
  • In a study of 10 VEXAS patients, two were found with Mycobacterium avium, alongside a review of 20 cases of disseminated NTMi over 13 years, highlighting common conditions among the patients.
  • The findings suggest that VEXAS patients may have a higher susceptibility to NTMi due to issues with their immune response, specifically monocytic dysfunction, and emphasize the need for vigilant diagnosis of opportunistic infections before adjusting

  • The presence of donor Vγ9Vδ2 T-cells after haploidentical hematopoietic stem cell transplant (h-HSCT) is linked to better disease-free survival, as these cells can target tumor cells without triggering severe side effects.
  • A clinical study assessed the safety of generating these T-cells by combining zoledronic acid (ZA) with interleukin-2 (IL-2), showing that the procedure is safe and successfully increases T-cell numbers in patients.
  • The study's outcomes suggest potential for further research into using this immunotherapy to reduce relapse rates after h-HSCT, indicating that more extensive trials may come next.
