YY1 is a C2H2-type zinc finger transcription factor that is a member of the human GLl-Kruppel family of proteins. YY1 represses transcription when bound upstream of transcription initiation sites. The repression can be relieved by adenovirus E1A and activation of target genes occurs.

Cochlear implants for congenitally deaf adolescents: is open-set speech perception a realistic expectation?

The prognosis for benefit from use of cochlear implants in congenitally deaf adolescents, who have a long duration of profound deafness prior to implantation, has typically been low. Speech perception results for two congenitally deaf patients implanted as adolescents at the University of Melbourne/Royal Victorian Eye and Ear Hospital Clinic show that, after 12 months of experience, both patients had significant open-set speech discrimination scores without lipreading. These results suggest that although benefits may in general be low for congenitally deaf adolescents, individuals may attain significant benefits to speech perception after a short period of experience.

Comparison of three methods for measuring cigarette smoking in patients with vascular disease.

Although smoking has been identified as an independent risk factor in peripheral vascular disease, smokers do not accurately report their habit. Therefore, a cheap, non-invasive, objective method of measuring smoking behaviour in vascular surgical units is desirable. The value of end-expiratory carbon monoxide monitoring, which is both simple and inexpensive, was compared with two well-established, validated techniques (serum thiocyanate and blood carboxyhaemoglobin levels).

Retinoblastoma and p53 tumor suppressor genes in human hepatoma cell lines.

We analyzed the status of retinoblastoma and p53 genes in 10 human hepatoma cell lines. Polyclonal anti-peptide antibodies generated against peptides homologous to COOH-terminal and leucine-zipper domains of the retinoblastoma protein allowed us to identify two cell lines (Hep 3B and FOCUS) with abnormal expression. The same cell lines have both lacked p53 expression.

Evidence for physical interaction between the zinc-finger transcription factors YY1 and Sp1.

Two promoter elements are important for basal-level transcription, the TATA motif typically located 30 nucleotides upstream of the transcription initiation site and the initiator (Inr) element encompassing the start site. The mechanism of how Inr elements work is poorly understood, partly because very few proteins that bind to Inr elements have been identified and isolated. The recently cloned YY1 is such an Inr-binding protein.

Factors in the development of a training program for use with tactile devices.

A review of the literature suggests that, in order to maximize the benefits available through a tactile device, it must be accompanied by an effective and adaptive training program. There are a number of factors to consider in the design of such a training program, including the type of tasks and response formats to include, the amount of training, subject motivation and device use, the characteristics of the potential user population, the specific device to be used and the type of information it provides, and the evaluation procedures to be followed. The type and saliency of the information provided by a particular tactile device are highlighted as the most important yet neglected consideration in designing a training program.

The effect of handedness in tactile speech perception.

This study examined differential performance of normally hearing subjects using a tactile device on the dominant versus non-dominant hand. The study evaluated whether tactual sensitivity for non-speech stimuli was greater for the dominant hand as compared with the non-dominant hand, and secondly, whether there was an advantage for speech presented tactually to the dominant hand, resulting from a preferential pathway to the language processing area in the left cerebral hemisphere. Evaluations of threshold pulse width, dynamic ranges, paired electrode identification, and a closed-set tactual pattern discrimination test battery showed no difference in tactual sensitivity measures between the two hands.

The major histocompatibility complex class I antigen-binding protein p88 is the product of the calnexin gene.

A 90-kDa phosphoprotein (p90) of the endoplasmic reticulum was identified by a monoclonal antibody generated against human hepatoma cells. Pulse-chase experiments with [32P]phosphate and [35S]methionine demonstrated that p90 formed both stable and transient complexes with other cellular proteins, suggesting its role as a molecular chaperone. This protein associates with heavy chains of major histocompatibility complex class I proteins, suggesting that it is the human homolog of the recently described 88-kDa protein that transiently associates with murine class I molecules in the endoplasmic reticulum.

Identification of polymorphic simple sequence repeats in the genome of the zebrafish.

The zebrafish has drawn a great deal of attention as a developmental system because it offers the ability to combine excellent embryology and genetics. Here, we report that simple sequence repeats are abundant in the zebrafish genome and are highly polymorphic between two outbred lines, making them useful markers for the construction of a genetic map of this organism.

Nutritional ecology of pastoralists in dry tropical Africa.

African pastoralists inhabit a variety of environments within tropical arid to semiarid regions. Ecological and cultural factors as well as economic opportunities and constraints influence the amounts and types of foods available. The preferred staple of all pastoral populations is milk from livestock.

Teicoplanin vs cephradine and metronidazole in the prophylaxis of sepsis following vascular surgery: an interim analysis of an ongoing trial.

This paper presents further preliminary results of a trial of the prophylaxis of sepsis in 165 patients undergoing vascular surgery. The efficacy and safety of a single dose of teicoplanin was examined and compared with three doses of cephradine plus metronidazole. No significant differences were detected in the prophylactic efficacy in either group.

Use of a multichannel electrotactile speech processor by profoundly hearing-impaired children in a total communication environment.

As part of a larger subject group, four profoundly hearing-impaired children enrolled in a total communication educational program were fitted with the University of Melbourne's multichannel electrotactile speech processor (Tickle Talker). Sound detection thresholds for pure tones were at lower levels with the tactile device than with hearing aids, especially for high frequency sounds above 2 kHz. Two of the children also detected all speech sounds of the Ling five-sound test at normal conversational levels using only the Tickle Talker.

Role of a multichannel electrotactile speech processor in a cochlear implant program for profoundly hearing-impaired adults.

Four profoundly hearing-impaired adults who did not meet current selection criteria for implantation at the University of Melbourne were each fitted with a wearable multichannel electrotactile speech processor (Tickle Talker). The subjects were evaluated with a test battery of speech discrimination tests subsequent to training in use of the device. Thresholds for detection of pure tones were lower for the Tickle Talker than for hearing aids across the frequency range 250 to 4000 Hz.

Perception of multiple electrode stimulus patterns: implications for design of an electrotactile speech processor.

Identification of multiple-electrode stimulus patterns was evaluated in nine adult subjects, to assess the feasibility of providing additional speech information through the tactual display of an electrotactile speech processor. Absolute identification scores decreased from 97.8% for single electrodes, to 61.

Variability in protection afforded by half-mask respirators against styrene exposure in the field.

Concentrations of styrene were measured in the breathing zone and inside the facepiece of air-purifying half-mask respirators of 13 workers for three to six 1-hr periods while they were engaged in the production of fiberglass-reinforced products. These data were used to estimate the protection afforded by the respirators. In arriving at these estimates, it was necessary to correct the concentration measured inside the mask for pulmonary retention of styrene by the workers.

Perception of amplitude envelope variations of pulsatile electrotactile stimuli.

Gross variations of the speech amplitude envelope, such as the duration of different segments and the gaps between them, carry information about prosody and some segmental features of vowels and consonants. The amplitude envelope is one parameter encoded by the Tickle Talker, an electrotactile speech processor for the hearing impaired which stimulates the digital nerve bundles with a pulsatile electric current. Psychophysical experiments measuring the duration discrimination and identification, gap detection, and integration times for pulsatile electrical stimulation are described and compared with similar auditory measures for normal and impaired hearing and electrical stimulation via a cochlear implant.

Perception of sentences, words, and speech features by profoundly hearing-impaired children using a multichannel electrotactile speech processor.

Fourteen prelinguistically profoundly hearing-impaired children were fitted with the multichannel electrotactile speech processor (Tickle Talker) developed by Cochlear Pty. Ltd. and the University of Melbourne.

Enhanced expression of the protein kinase substrate p36 in human hepatocellular carcinoma.

A basic phosphoprotein defined by a monoclonal antibody named AF5 was found to be highly abundant in human hepatocellular carcinoma by Western immunoblotting. Under the same conditions, the levels of this phosphoprotein were low or undetectable in normal liver extracts. The AF5 antibody was used to screen a cDNA expression library of a human hepatoma cell line named FOCUS.

Abnormal structure and expression of p53 gene in human hepatocellular carcinoma.

There is little information regarding the molecular mechanisms of hepatocarcinogenesis. We studied the p53 gene at the DNA, RNA, and protein level in seven human hepatocellular carcinoma (HCC)-derived cell lines; six of seven showed p53 abnormalities. By Southern blotting, the p53 gene was found to be partially deleted in Hep 3B and rearranged in SK-HEP-1 cells.

A comparison of teicoplanin vs. cephradine and metronidazole in surgical prophylaxis: an interim analysis.

The preliminary results of a trial to examine and compare the safety, tolerability and efficacy of a single dose of teicoplanin with three doses of cephradine combined with metronidazole are presented in a series of 113 patients undergoing elective vascular surgery. There were no obvious differences in the infection rates and sepsis indicators in either group. Neither drug regimen produced any evidence of renal or hepatic damage, though the levels of three hepatocellular enzymes, aspartate aminotransferase (AST), alanine aminotransferase (ALT) and gamma-glutamyl-transferase (GGT), were seen to be transiently elevated, peaking 7 days post-operatively.

Intravenous oxypentifylline in critical limb ischaemia.

In an open tolerability study, 36 consecutive patients with critical ischaemia of the lower limbs were treated with intravenous oxypentifylline 1, 200 mg daily for up to three weeks. Patients were assessed objectively using a radioisotope limb blood flow technique and a subjective assessment of the patient's pain was made using linear analogue pain scores and analgesia scores. Treatment was well tolerated in 34 patients but was discontinued in two because of significant side effects.
