The hypotheses that intrahepatic bile ducts are derived either by a transformation of periportal liver cells or by dichotomous branching of the extrahepatic bile ducts were investigated in fetal and postnatal rat livers by histological and immunohistochemical methods using an antiserum to prekeratin which, in the liver, binds to biliary epithelial cells (BEC). In conventionally stained sections, bile duct development was observed to begin in the 19 day fetus around the larger branches of the portal vein, with the formation of lumina surrounded by cuboidal or elongated hepatoblast-like cells on the portal aspect and readily distinguished hepatoblasts on the lobular aspect. At 21 days, these structures had developed into canals of Hering lined jointly by recognizable liver cells and BEC.

We have isolated a cDNA clone (SE5A) that encodes a protein on lampbrush chromosome loops of the newt Notophthalmus. In vitro-synthesized transcripts of this clone were injected into Xenopus oocytes, where they were efficiently translated. Most of the translated protein was imported into the oocyte nucleus, and some of it appeared on the chromosome loops.

The proposal that the cytoplasmic granules of glomerular peripolar cells (PPCs) of the adult sheep and lamb contain a kallikrein-like substance has been re-examined. After extensive characterisation and purification of anti-kallikrein antibodies, it was concluded that PPCs do not contain immunohistochemically detectable amounts of urinary kallikrein; nor do such cells appear to synthesise kallikrein as shown by hybridisation histochemistry studies. They are also not renin-immunopositive.

The authors studied the H ferritin restriction polymorphism in 83 hemochromatosis patients and 84 controls as well as in 19 nuclear families. No significant difference was found with the ten restriction enzymes used (HindIII, EcoRI, EcoRV, PvuII, BamHI, PstI, Bg/I, Bg/II, HincII, and TaqI). Hence, the genomic abnormality responsible for idiopathic hemochromatosis is not a major deletion of an H ferritin gene.

The genes coding for the large and small subunits of the periplasmic hydrogenase from Desulfovibrio baculatus have been cloned and sequenced. The genes are arranged in an operon with the small subunit gene preceding the large subunit gene. The small subunit gene codes for a 32 amino acid leader sequence supporting the periplasmic localization of the protein, however no ferredoxin-like or other characteristic iron-sulfur coordination sites were observed.

The hydrogenase from Desulfovibrio baculatus (DSM 1743) was purified from each of three different fractions: soluble periplasmic (wash), soluble cytoplasmic (cell disruption) and membrane-bound (detergent solubilization). Plasma-emission metal analysis detected in all three fractions the presence of iron plus nickel and selenium in equimolecular amounts. These hydrogenases were shown to be composed of two non-identical subunits and were distinct with respect to their spectroscopic properties.

A method for the isolation and culture of intact intrahepatic bile ducts from normal rats, and its use in studying putative inducers of biliary epithelial cell (BEC) hyperplasia was developed. Ducts were isolated by sequential perfusion of the liver with EGTA and collagenase-hyaluronidase followed by mild mechanical agitation. The resultant fraction, consisting of numerous small bile ducts within a connective tissue framework, was collected and embedded in a collagen gel and cultured on a raft assembly in Medium 199 supplemented with 15% newborn calf serum and antibiotics.

Satellite 2 of the newt, Notophthalmus viridescens, is a 330 bp tandemly repeated sequence scattered throughout the genome. Cytoplasmic transcripts homologous to satellite 2 are found in a variety of tissues. Most of the transcripts correspond precisely in length to the DNA repeat unit or to whole multiples of that repeat.

We have investigated transcription of the histone gene cluster on lampbrush chromosomes of the newt Notophthalmus viridescens. Clusters of the five histone genes are separated by long tracts of a DNA repeat called satellite 1. Previous in situ hybridization results demonstrated the presence of histone gene coding regions, intergene spacers, and flanking satellite repeats in giant transcription units on lampbrush chromosomes.

Details are given of a technique for making permanent preparations of the lampbrush chromosomes of Xenopus laevis. Stained preparations allow all 18 bivalent chromosomes to be identified, and a working map showing the major features has been constructed. Fifteen of the Xenopus chromosomes have one telomere conspicuously larger than the other; the two smallest chromosomes, and one other, lack large telomeres.
