A simple array platform for microRNA analysis and its application in mouse tissues.

MicroRNAs (miRNAs) are a novel class of small noncoding RNAs that regulate gene expression at the post-transcriptional level and play a critical role in many important biological processes. Most miRNAs are conserved between humans and mice, which makes it possible to analyze their expressions with a set of selected array probes. Here, we report a simple array platform that can detect 553 nonredundant miRNAs encompassing the entire set of miRNAs for humans and mice.

Membrane topology of the GltS Na+/glutamate permease of Escherichia coli.

The GltS Na+/glutamate permease of Escherichia coli is the most extensively studied member of the ESS family of bacterial glutamate:Na+ symporters. This paper presents the membrane topology analysis of the GltS with translational alkaline phosphatase and beta-galactosidase gene fusions generated by TnphoA, nested deletions and targeted fusions. The topology model suggested by the translational fusion technique is compared with the MemGen model and discussed in detail.

Bioavailability of arsenic and antimony in soils from an abandoned mining area, Glendinning (SW Scotland).

The mobility and bioavailability of As and Sb in relation to soil-biota transfer were evaluated at a former Sb mining and smelting site (Glendinning, SW Scotland, UK). The study specifically assessed the accumulation of As and Sb in different environmental components (soil, plants and earthworms) across mining area to estimate risk factors for the biota. Total concentrations and fractions of As and Sb in soils were determined.

Effect of massage of the hamstring muscles on selected electromyographic characteristics of biceps femoris during sub-maximal isometric contraction.

The aim of this study was to investigate whether a single massage of the hamstring muscles would alter selected electromyographic characteristics of biceps femoris during a sub-maximal isometric contraction. Eleven healthy young males participated in this crossover study. They were randomly assigned to two groups, receiving either a 15-minute hamstring muscle massage or a 15-minute prone rest.

Expression and protective role of heme oxygenase-1 in delayed myocardial preconditioning.

In the study the authors aimed to demonstrate the expression and protective effect of heme oxygenase-1 (HO-1) in the delayed preconditioning (PC) on cultured myocardiac cells. Neonatal rat cardiac myocytes were exposed to ischemic (ischemic medium [IM] for 20 min) and pharmacological (adenosine, epinephrine, opioid) PC. Twenty-four hours later cells were subjected to a simulated ischemia (SI)--culturing for 3 h in IM, followed by 2-h reperfusion in normal medium--and then lactate dehydrogenase (LDH), live/death ratio, and apoptosis were measured.

Cardioprotective action of urocortin in early pre- and postconditioning.

Pre- and postconditioning are powerful endogenous adaptive phenomenon of the organism whereby different stimuli enhance the tolerance against various types of stress. Urocortin (Ucn), member of the corticotropin-releasing factor (CRF) family has potent effects on the cardiovascular system. The aim of this article was to investigate the action of Ucn on cultured cardiomyocytes in the process of pre- and postconditioning.

Metal uptake by woodlice in urban soils.

This study evaluated the uptake of bioavailable metal (Cd, Cr, Cu, Fe, Pb and Zn) by woodlice (Isopoda) collected from public open spaces in urban areas of Renfrewshire, Central West Scotland, UK. The species Oniscus asellus and Porcellio scaber were collected at 13 different locations together with associated surface soil samples. Soils were subject to sequential extraction to evaluate metal availability and analyzed by ICP-AES and flame AAS for Cd, Cr, Cu, Fe, Pb and Zn concentrations.

Coronary revascularization in transition from on-pump to off-pump: the effect of the off-pump coronary artery bypass on medium-term outcome.

Aim: We previously reported that early patient outcome, chiefly ischaemic injury, was reduced in patients allocated to off pump coronary artery bypass (OPCAB) surgery. This report concerns the medium-term outcome for this cohort of patients.

Methods: A prospective observational study was carried out in a single cardiothoracic specialty hospital.

p62 accumulates and enhances aggregate formation in model systems of familial amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a progressive neurode-generative disease characterized by motor neuron death. A hallmark of the disease is the appearance of protein aggregates in the affected motor neurons. We have found that p62, a protein implicated in protein aggregate formation, accumulated progressively in the G93A mouse spinal cord.

Long-term outcome of patients with perioperative myocardial infarction as diagnosed by troponin I after routine surgical coronary artery revascularization.

Objective: Diagnosis of perioperative myocardial infarction (P-MI) after coronary artery bypass graft (CABG) surgery traditionally relied on a combination of electrocardiographic and enzyme assay changes. Patients with Q-wave P-MIs who survive to hospital discharge have a poorer long-term prognosis. Troponin assays are more sensitive and specific for detecting minor P-MI, with an increased incidence of P-MI being reported.

The influence of training with heavy rugby balls on selected spin pass variables in youth rugby union players.

The purpose of this study was to investigate the influence of training with heavy rugby balls on selected spin pass variables in youth rugby union players. Pre-training, 14 participants performed rapid spin passes (using right and left hands) at a wall-mounted target, from 7-m and 10-m distances. Horizontal (linear) and spin (longitudinal angular) velocities of the passes were recorded using a 25Hz video camera.

Carl Friedrich Naumann and the introduction of enantio terminology: a review and analysis on the 150th anniversary.

Enantiomorphism and enantiomorphous were the first enantio-based terms, introduced 150 years ago, by Carl Friedrich Naumann, a German crystallographer, to refer to non-superposable mirror-image crystals. The terminology was not adopted by Pasteur, the discoverer of molecular chirality, and was not embraced at first in the stereochemical context, until it was accepted in 1877 by Van't Hoff in the German edition of his proposal for the tetrahedral asymmetric carbon atom. In the 1890s the use of enantio terms began to spread in the research literature, and many new derivatives of Naumann's original two terms were subsequently introduced.

Gas-phase protonation and deprotonation of acrylonitrile derivatives N[triple chemical bond]C--CH==CH--X (X=CH(3), NH(2), PH(2), SiH(3)).

A combined experimental and theoretical study on the gas-phase basicity and acidity of a series of cyanovinyl derivatives is presented. The gas-phase basicities and acidities of (N[triple chemical bond]C--CH==CH--X, X=CH(3), NH(2)) were obtained by means of Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry techniques. The corresponding calculated values were obtained at the G3B3 level of theory.

Computational determination of refractive index distribution in the crystalline cones of the compound eye of Antarctic krill (Euphausia superba).

In order to understand how a compound eye channels light to the retina and forms an image, one needs to know the refractive index distribution in the crystalline cones. Direct measurements of the refractive indices require sections of fresh, unfixed tissue and the use of an interference microscope, but frequently neither is available. Using the eye of the Antarctic krill Euphausia superba (the main food of baleen whales) we developed a computational method to predict a likely refractive index distribution non-invasively from sections of fixed material without the need of an interference microscope.

Complexes between lithium cation and diphenylalkanes in the gas phase: the pincer effect.

The gas-phase lithium cation basicities (LCB values, Gibbs free energies of binding) of alpha,omega-diphenylalkanes Ph-(CH(2))(n)-Ph (n=2, 3, or 7) and 1,1-diphenylethane Ph-CH(Me)-Ph were investigated by means of Fourier-transform ion cyclotron resonance (FTICR) mass spectrometry. Their structures, and those of the corresponding Li(+) complexes were optimized at the B3LYP/6-31G(d) level and their relative stabilities calculated at the B3LYP/6-311+G(3df,2p)//B3LYP/6-31G(d) level. Whereas the most stable conformers of the free diphenylalkanes were found to adopt a completely stretched aliphatic chain connecting the two benzene rings, the most stable Li(+) complexes correspond to conformers in which the alkali metal cation interacts simultaneously with both benzene rings through the folding of the aliphatic chain ("pincer effect").

Effect of cardiopulmonary bypass on cytokine network and myocardial cytokine production.

Background: In addition to the well-investigated proinflammatory cytokine expression, there is an ever increasing interest in the field of anti-inflammatory response to cardiopulmonary bypass (CPB). Evidence suggests that myocardium serves as an important source of cytokines during reperfusion and application of CPB. The effect of coronary artery bypass graft (CABG) without CPB on myocardial cytokine production has not as yet been investigated.

Pre-operative coronary revascularization: an optimal therapy for high-risk vascular surgery patients?

Cardiac complications are the leading cause of peri-operative morbidity and mortality of patients undergoing vascular surgery. This high incidence of cardiac complications is related to the presence of underlying coronary artery disease. The optimal treatment strategy for these high-risk patients, including the use of pre-operative coronary revascularization for the purpose of improving peri-operative and long-term cardiac outcomes, has been controversial for several decades.

Identification of essential host factors affecting tombusvirus RNA replication based on the yeast Tet promoters Hughes Collection.

To identify essential host genes affecting replication of Tomato bushy stunt virus (TBSV), a small model plant virus, we screened 800 yeast genes present in the yeast Tet promoters Hughes Collection. In total, we have identified 30 new host genes whose down-regulation either increased or decreased the accumulation of a TBSV replicon RNA. The identified essential yeast genes are involved in RNA transcription/metabolism, protein metabolism/transport, or other cellular processes.

Experimental and theoretical study of skylight polarization transmitted through Snell's window of a flat water surface.

The celestial polarization pattern may be scrambled by refraction at the air-water interface. This polarization pattern was examined in shallow waters with a submersible polarimeter, and it was calculated by using land measurements ('semiempirical predictions') and models of the skylight polarization. Semiempirically predicted and measured e-vector orientations were significantly similar.

Ab initio and experimental thermodynamic and kinetic study of proton-assisted bond activation in gaseous hydrocarbons: deconvolution of reaction efficiencies in the case of adamantane.

1) Protonation at all possible sites of adamantane (C(10)H(16)) was studied at the MP2/6-311++G(3df,2p)//MP2/6-311++G(d,p) level. This provided values of the changes in the thermodynamic state functions for these processes. Whenever direct comparison was possible, the agreement with experimental data was very good.

Bonding energetics in clusters formed by cesium salts: a study by collision-induced dissociation and density functional theory.

In relation to the interaction between (137)Cs and soil organic matter, electrospray mass spectrometry experiments and density functional theory (DFT) calculations were carried out on the dissociation of positively charged adducts formed by cesium nitrate and cesium organic salts attached to a cesium cation [Cs(CsNO(3))(CsA)](+) (A = benzoate, salicylate, hydrogen phthalate, hydrogen maleate, hydrogen fumarate, hydrogen oxalate, and hydrogen malonate ion). These mixed clusters were generated by electrospray from methanol solutions containing cesium nitrate and an organic acid. Collision-induced dissociation of [Cs(CsNO(3))(CsA)](+) in a quadrupole ion trap gave [Cs(CsNO(3))](+) and [Cs(CsA)](+) as major product ions.

Cardiac risk reduction in non-cardiac surgery: the role of anaesthesia and monitoring techniques.

Cardiac complications are the major cause of perioperative morbidity and mortality of patients undergoing non-cardiac surgery. This is related to the frequent presence of underlying coronary artery disease. In the last few decades, attention has focused on preoperative cardiac risk assessment that may help to identify patients at increased cardiac risk for whom cardioprotective medication and, when indicated, coronary revascularization may improve perioperative outcome.

[Risk of cardiac complications in patients undergoing major vascular surgery].

In patients undergoing major vascular surgery cardiac complications are the major cause of perioperative morbidity and mortality. This is related to the frequent presence of underlying coronary artery disease. The aim of the preoperative cardiac risk assessment is to evaluate the presence and degree of coronary artery disease along with other risk factors such as cerebrovascular disease, renal insufficiency, and diabetes mellitus that may influence the perioperative risk of these patients.

Synthesis and structural characterization of new oxorhenium and oxotechnetium complexes with XN2S-tetradentate semi-rigid ligands (X = O, S, N).

Twelve novel oxo-technetium and oxo-rhenium complexes based on N2S2-, N2SO- or N3S-tetradentate semi-rigid ligands have been synthesised and studied herein. By reacting the ligands with a slight excess of suitable [MO]3+ precursor (ReOCl3(PPh3)2 or [NBu4][99gTcOCl4]), the monoanionic complexes of general formula [MO(Ph-XN2S)]- could be easily produced in high yield. The complexes have been characterized by means of IR, electrospray mass spectrometry, elemental analysis, NMR and conductimetry.

Molecular and morphological heterogeneity of neural precursors in the mouse neocortical proliferative zones.

The proliferative ventricular zone (VZ) is the main source of projection neurons for the overlying cerebral neocortex. The number and diversity of neocortical neurons is determined, in part, by factors controlling the proliferation and specification of VZ cells during embryonic development. We used a variety of methods, including in utero electroporation with specific cellular markers, computer-assisted serial EM cell reconstruction, and time-lapse multiphoton imaging to characterize the molecular and morphological characteristics of the VZ constituents and to capture their behavior during cell division.
