1H double-quantum and COSY NMR-spectra of the porcine protein adenylate kinase have been obtained in order to assign aromatic spin systems within this protein, which contains seven tyrosyl, five phenylalanyl, and two histidyl residues. Double-quantum spectroscopy in its simple form allowed the assignment of five connectivities in the aromatic region and therefore proved inferior to COSY, which allowed the assignment of connectivities within nine aromatic residues.

In unequal alternating monocular exposure, each eye receives normal patterned input, but on alternate days and for unequal periods. This imbalance in stimulation produces a behavioral deficit for the less-experienced eye and alters the ability of that eye to activate cortical cells. To determine whether unequal alternating exposure also affects the sizes of cells in the lateral geniculate nucleus (LGN), we measured the cross-sectional areas of geniculate neurons in seven normally reared cats, 14 cats reared with equal alternating exposure, and 17 cats reared with unequal alternating exposure.

Tricyclic antidepressants (TADs) are known to antagonize the hypotensive and sedative actions of clonidine. We compared the effects of bupropion and imipramine pretreatment on the acute hypotensive and sedative actions of clonidine in eight normotensive male subjects in a randomized, double-blind crossover study. Pretreatment with bupropion, 100 mg by mouth three times a day for 9 days, had no effect on baseline supine blood pressure (BP) and heart rate (HR) and did not modify the hypotensive, bradycardic, and sedative actions of clonidine.

The gamma-subunit of mouse 7S nerve growth factor (gamma-NGF) is a member of a family of closely related serine proteases that includes kallikreins and tamases. We have isolated from a DBA/2J male submaxillary gland cDNA library a clone, pSM676, which codes for gamma-NGF. Sequence analysis of the clone shows that it codes for the C-terminal 138 amino acids of the protein plus 23 bases of the 3'-nontranslated portion of the message.

We report the construction of a small library of recombinant plasmids containing Mus musculus repetitive DNA inserts. The repetitive cloned fraction was derived from denatured genomic DNA by reassociation to a Cot value at which repetitive, but not unique, sequences have reannealed followed by exhaustive S1 nuclease treatment to degrade single stranded DNA. Initial characterizations of this library by colony filter hybridizations have led to the identification of a previously undetected M.

A technique suitable for measuring fluctuating temperatures in supersonic turbulent flows of N(2) seeded with NO has been demonstrated in a nonflowing cell. The method relies on the two-photon excitation of two selected rovibronic transitions in the NO gamma(A(2)Sigma+,'=0?X(2)II,''=0) band. Measurements between 155 and 295 K in N(2) mixtures containing 300 parts in 10(6) NO were obtained with single-pulse errors below 4% rms.

Seven tk- mutants of herpes simplex virus, type 2 (HSV-2), and three tk- mutants of herpes simplex virus, type 1 (HSV-1), were isolated which did not produce the thymidine kinase (TK) polypeptides but formed smaller polypeptides not seen in wild-type infected cells. Positive TK mRNA selection by hybridization to the cloned tk genes followed by in vitro translation identified the TK polypeptides. Comparisons of the products of partial proteolysis of the polypeptides of four HSV-2 and two HSV-1 tk- mutants to those of the parental TK polypeptides indicated that, in each case, the novel polypeptide was a fragment of the TK polypeptide, showing that these mutants have defects in the structural gene for tk.

We have constructed recombinant DNA clones containing small complementary DNA (cDNA) sequences homologous to portions of a 4.8-kb yeast viral double-stranded RNA (dsRNA) (L1) that codes for the viral capsid polypeptide. Neither the viral dsRNA nor its in vitro transcript is polyadenylated; hence the cDNAs were synthesized by reverse transcriptase on the in vitro mRNA transcript made by the viral transcriptase, using sheared salmon sperm DNA as a random primer.

The renin regulatory locus (Rnr) is a genetic element governing mouse submaxillary gland (SMG) renin levels. A 45,000 dalton polypeptide detectable after in vitro translation of mouse SMG mRNA has been identified by genetic and physical criteria as SMG renin. A cDNA recombinant clone specific for SMG renin has been isolated and used to demonstrate that the previously described genetic regulation of SMG renin levels is manifest at the level of renin mRNA concentration.

The intrathecal administration of beta-endorphin in the primate through an indwelling spinal catheter, produced a significant elevation in the nociceptive threshold as measured by the discrete trial shock titration task. The time of onset, duration of effect and magnitude of effect were all dose-dependent over a range of 150-750 micrograms. The effects were antagonized in a dose-dependent fashion by the systemic administration of naloxone.

Nutritional variant streptococci identified as pyridoxal-dependent Streptococcus mitior (mitis) account for 5 to 6% of streptococcal endocarditis and may be a cause of "culture-negative" endocarditis. Hence, growth of three variant strains in 11 commercial blood culture broths was compared to that in fresh heart infusion broth. For simulation of clinical specimens, culture bottles were injected with 5 ml of human blood, inoculated with approximately 500 colony-forming units (CFU) per bottle, and monitored for 7 days with Gram stains and viable counts.

A laser-induced fluorescence technique, especially suitable for measuring fluctuating temperatures in cold turbulent flows containing very low concentrations of nitric oxide, is described and analyzed. Temperatures below 300 K may be resolved with SNRs of >50:1, using commercially available high-peak-power tunable dye lasers. The purpose of this work was to assess the potential effect that chronic inhalation of diesel exhaust may have on lung mechanics and lung volume. Noninvasive pulmonary function tests that produced data on lung air flows and volumes have been conducted repeatedly on 25 male Fischer-344 rats exposed to diesel exhaust at a particulate concentration of 1500 micrograms m-3, 20 h per day, 5 1/2 days per week, for 612 days. The same tests were conducted on 25 clean air control animals.

The effects of a schedule of discontinuously negatively correlated reinforcement (DNC), in which only approach responses that take 5 sec or longer are reinforced, were studied in weanling rats that began training at 18 days of age and adolescents that began training at 60 days. Yoked partially reinforced subjects (PRF) received rewards on the same trials as corresponding DNC subjects of the same age, whereas continuously reinforced subjects (CRF) were rewarded on all trials. At the end of DNC training (210 trials), the older but not the younger DNC subjects were running slower than their yoked-PRF controls.
