The secretory capacity of Chinese hamster ovary (CHO) cells remains a fundamental bottleneck in the manufacturing of protein-based therapeutics. Unconventional biological drugs with complex structures and processing requirements are particularly problematic. Although engineered vector DNA elements can achieve rapid and high-level therapeutic protein production, a high metabolic and protein folding burden is imposed on the host cell.

The respiratory tract hosts a diverse microbial community whose composition varies with anatomical location and throughout life. Rothia mucilaginosa, a common commensal of the upper respiratory tract and oral cavity, has recently been recognized for its ability to inhibit bacteria-triggered pro-inflammatory responses. However, its role in modulating the immune response to viral infections such as influenza A virus (IAV) pneumonia, remains unknown.

Imaged capillary isoelectric focusing was successfully applied for separating an in-house synthesized closely related peptide pair, that is, a linear 12-mer (Rp5-L) and its cyclic 15-mer variant (Rp5-C). Rp5-L represents a mimotope, that is, an epitope mimicking peptide, of the CD20 antigen, which is over-expressed in B-cell-related tumors. Peptide identity-including the successful disulfide bond formation in Rp5-C-was confirmed with matrix-assisted laser desorption ionization-time of flight mass spectrometry.

A formerly developed mathematical model describing drug release from hydrophilic matrices (HMs) took into account resistance to drug release given by its dissolution and by the presence of a growing gel layer. Such a model was applied to previously reported release data obtained from HMs made of hydroxypropyl methylcellulose (HPMC), where acetaminophen was used as model drug and a cellulolytic product was added as "active" excipient to attain zero-order release kinetics. The Levich theory applied to acetaminophen intrinsic dissolution rate (IDR) data highlighted the suitability of such a drug for modeling purposes, given its good surface wettability.

Background/objectives: The present work proposes a mathematical model able to describe the dissolution of poly-disperse drug spherical particles in a solution (Dissolution Rate Test-DRT). DRT is a pivotal test performed in the pharmaceutical field to qualitatively assess drug bioavailability.

Methods: The proposed mathematical model relies on the key hallmarks of DRT, such as particle size distribution, solubility, wettability, hydrodynamic conditions in the dissolving liquid of finite dimensions, and possible re-crystallization during the dissolution process.

Article Synopsis
  • - Mechanical testing of articular cartilage shows high variability due to factors like sample geometry; our study focused on how sample shape impacts its mechanical response during unconfined compression tests.
  • - Using advanced imaging techniques and finite element modeling, we found that geometric irregularities in cartilage samples from a single bovine knee significantly affected their mechanical properties, including a 15% decrease in force due to an inclined shape.
  • - Our analysis revealed that accounting for these irregularities led to substantial changes in material parameters, with increases in stiffness and permeability and a considerable reduction in fitting error, highlighting the importance of sample geometry in cartilage testing.

Demoralization comprises multiple dimensions. Among them, Subjective Incompetence (SI) is the perception of being incapable of appropriate action in demanding circumstances. SI may be an early sign of demoralization preceding hopelessness, thus we aimed at integrating items related to Subjective Incompetence into the clinical assessment of demoralization.

One step towards understanding bone fragility and degenerative diseases is to unravel the links between fracture resistance and the compositional and structural characteristics of cortical bone. In this study, we explore an optical method for automatic crack detection to generate full fracture resistance curves of cortical bone. We quantify fracture toughness, critical failure strains at the crack tip, and crack tortuosity in three directions and analyze how they relate to cortical bone microstructure.

Background: The superimposed pressure is the primary determinant of the pleural pressure gradient. Obesity is associated with elevated end-expiratory esophageal pressure, regardless of lung disease severity, and the superimposed pressure might not be the only determinant of the pleural pressure gradient. The study aims to measure partitioned respiratory mechanics and superimposed pressure in a cohort of patients admitted to the ICU with and without class III obesity (BMI ≥ 40 kg/m), and to quantify the amount of thoracic adipose tissue and muscle through advanced imaging techniques.

Despite the need for effective treatments against chronic respiratory infections (often caused by pathogenic biofilms), only a few new antimicrobials have been introduced to the market in recent decades. Although different factors impede the successful advancement of antimicrobial candidates from the bench to the clinic, a major driver is the use of poorly predictive model systems in preclinical research. To bridge this translational gap, significant efforts have been made to develop physiologically relevant models capable of recapitulating the key aspects of the airway microenvironment that are known to influence infection dynamics and antimicrobial activity In this review, we provide an overview of state-of-the-art cell culture platforms and models that have been used to model chronic (biofilm-associated) airway infections, including air-liquid interfaces, three-dimensional cultures obtained with rotating-wall vessel bioreactors, lung-on-a-chips and pig lungs.

Objectives: Disclosing information on diagnosis, prognosis and treatment is a delicate process in oncology, although awareness levels have over time increased in people with cancer. However, individual characteristics should be considered when communicating difficult information. We conducted a multicentric study to explore the moderating role of coping styles on the relationship between information about cancer, quality of life and psychological distress.

The traditional youth-oriented design of Early Intervention Services (EIS) may lead to the exclusion of patients who have their psychotic onset later in life. A retrospective study was conducted to compare first-episode psychosis (FEP) patients who accessed treatment when aged ≤ 35 years with those ≥36+. A total of 854 patients were identified among 46,222 individuals who had access to community psychiatric services from 1991 to 2021.

The paucity of data regarding patients with Serious Mental Illness (SMI) and cancer is alarming given the fact that people with SMI, especially schizophrenia, bipolar disorders and severe depressive disorders, have in general poorer access to physical health care and higher morbidity and mortality because of physical illnesses. The aims of this review were to examine the current evidence from existing literature on the risk of developing cancer and its course among people with SMI. Equivocal results emerge regarding the risk of developing some kind of cancer among people with SMI, with contrasting data on a possible higher, similar or lower risk in comparison with the general population.

Article Synopsis
  • Electroconvulsive therapy (ECT) is a well-supported treatment for severe depression, but its neural mechanisms are not well understood; this review examines changes in cortical thickness (CT) due to ECT based on structural MRI studies.
  • Ten studies were analyzed, revealing that ECT significantly increases CT in brain regions like the temporal, insular, and frontal areas in patients with unipolar or bipolar depression; however, inconsistencies in sample sizes and study designs limit the applicability of the findings.
  • While the results suggest ECT may enhance CT and possibly affect treatment outcomes, further research with larger samples and better controls is needed to clarify the relationship between CT changes and clinical response.
