Sterile alpha motif and HD domain-containing protein 1 (SAMHD1) is a dNTP hydrolase important for intracellular dNTP homeostasis and serves as tumor suppressor and modulator of antimetabolite efficacy in cancer, though largely unexplored in breast cancer (BC). A cohort of patients with early BC (n = 564) with available gene expression data (GEP) was used. SAMHD1 protein expression was assessed by immunohistochemistry performed on tissue microarrays.

Article Synopsis
  • Stromal cells are crucial for maintaining the balance of epithelial and immune cells and are significant in the development of inflammatory bowel disease (IBD).
  • The research investigates the stromal response to inflammation in pediatric IBD, identifying specific inflammatory reactions in different parts of the colon and intestinal layers.
  • Findings show that certain fibroblasts and monocytes/macrophages interact closely in the intestine, with fibroblasts promoting the conversion of monocytes into a specific type of macrophage that resembles those found in young IBD patients, indicating the stroma's role in guiding macrophage development.

Autophagy is activated in response to a variety of stress conditions including anti-cancer therapies, and tumors cells often depend on autophagy for survival. In this study, we have evaluated inhibition of autophagy as therapeutic strategy in acute lymphoblastic leukemia (ALL) in children, both as a single treatment and in combination with glucocorticoid (GC) Dexamethasone (Dexa). Analysis of proteomics and RNA-seq of ALL cell lines and primary samples identified an upregulation of Vps34 and ATG14 proteins and autophagy and lysosomal pathway enrichment in a genetic subgroup with a recurrent t(12;21) translocation.

Background: Bronchopulmonary Dysplasia (BPD) in infants born prematurely is a risk factor for chronic airway obstruction later in life. The distribution of T cell subtypes in the large airways is largely unknown.

Objective: To characterize cellular and T cell profiles in the large airways of young adults with a history of BPD.

Sterile alpha motif and histidine-aspartate (HD) domain-containing protein 1 (SAMHD1) is a deoxynucleoside triphosphate triphosphohydrolase with ara-CTPase activity that confers cytarabine (ara-C) resistance in several hematological malignancies. Targeting SAMHD1's ara-CTPase activity has recently been demonstrated to enhance ara-C efficacy in acute myeloid leukemia. Here, we identify the transcription factor SRY-related HMG-box containing protein 11 (SOX11) as a novel direct binding partner and first known endogenous inhibitor of SAMHD1.
