The paper describes results of experimental studies of changes in the refractive index of the live cell (human erythrocyte) substance. The measurements were carried out using automated interference microscope. Oscillations of the refractive index were detected with the amplitude 5-10% and period 1 x 10(3)-3 x 10(3).

It was shown that the method has some advantages over the methods based on measurements of the intensity of light passing through a crossed polarizer, an analyzer, and a fiber (light polarized microscopy). The method involves direct phase measurements of optical path length at the parallel and perpendicular orientations of the polarization plane of probing radiation.

A device based on the Linnic microscope was constructed to obtain phase images, which are two-dimensional pictures of birefringence. For the first time, two-and-three-dimensional maps of both total birefringence and birefringence for individual sarcomeres in the central part of muscle fiber were visualized using large databases.
