Article Synopsis
  • This study investigates how Traditional Chinese Medicine (TCM) extracts can block the interaction of the SARS-CoV-2 Spike protein with the human ACE2 receptor, using advanced biochemical techniques.
  • The research involved testing 91 different TCM extracts, finding that Rosmarinus officinalis (rosemary) was the most effective at preventing viral entry and disrupting the Spike-ACE2 interaction.
  • The findings suggest that TCM extracts could be valuable in developing antiviral treatments, highlighting the potential of combining traditional methods with modern science.

Article Synopsis
  • The study explored how the fungal enzyme TadA synthesizes fusicoccane-type diterpenes by examining its cyclic formation process.
  • Extensive isotopic labeling experiments were conducted to gain a better understanding of this mechanism.
  • The research also looked into how the acidity (pH) levels affect the types of products TadA produces, revealing new insights into its functionality.

Article Synopsis
  • Fusicoccane (FC)-type diterpenoids have a unique 5-8-5 ring structure and show various biological activities; a new enzyme, MgMS, was discovered that produces a specific diterpene hydrocarbon skeleton.
  • Two additional gene clusters, Bn and Np, were found to encode similar terpene synthases to MgMS, resulting in the same product when expressed.
  • Introducing P450 enzymes from these clusters led to the creation of four new FC-type diterpenoids, with MgP450 and NpP450 responsible for significant hydroxylation and oxidation processes on the diterpene skeleton.

The diterpene synthase AfAS was identified from Aspergillus fumigatiaffinis. Its amino acid sequence and-according to a structural model-active site architecture are highly similar to those of the fusicocca-2,10(14)-diene synthase PaFS, but AfAS produces a structurally much more complex diterpene with a novel 6-5-5-5 tetracyclic skeleton called asperfumene. The cyclisation mechanism of AfAS was elucidated through isotopic labelling experiments and DFT calculations.

Article Synopsis
  • - Oxabornyl polyenes are a special type of polyketide found in fungi, featuring a distinct polyene core and diverse oxygen-containing rings, with known biological activities but complex configurations.
  • - The study isolated three specific oxabornyl polyenes (prugosenes A1-A3) from a fungal strain and used ozonolysis and ECD calculations to determine their absolute configurations, which were previously unassigned.
  • - The prugosenes showed strong antiviral effects against the respiratory syncytial virus, with a notable effectiveness of 6.3 μM for one of the compounds, contributing to a better understanding of these compounds and their potential uses.

Fernane-type triterpenoids are a small group of natural products mainly found in plants and fungi with a wide range of biological activities. Polytolypin is a representative fernane-type triterpenoid from fungi and possesses potent antifungal activity. So far, biosynthesis of fungal-derived fernane-type triterpenoids has not been characterized, which hinders the expansion of their structural diversity using biosynthetic approaches.

Article Synopsis
  • Fusicoccane-type terpenoids are a specialized group of diterpenoids known for their unique 5-8-5 ring structure and various biological activities, but only five synthesizing enzymes are currently identified.
  • A new biosynthetic gene cluster from the strain ATCC 26942 has been discovered, which includes a novel fusicoccane-type diterpene synthase gene, naming it TadA.
  • Experiments show that TadA creates a new compound, talaro-7,13-diene, using a different cyclization process, while another enzyme, TadB, oxidizes this compound into talaro-6,13-dien-5,8-dione.

Rock mass condition assessment during tunnel excavation is a critical step for the intelligent control of tunnel boring machine (TBM). To address this and achieve automatic detection, a visual assessment system is installed to the TBM and a lager in-situ rock mass image dataset is collected from the water conveyance channel project. The rock mass condition assessment task is transformed into a fine-grain classification task.
