The C3 protein is the central molecule within the complement system and undergoes proteolytic activation to C3b in the presence of pathogens. Pattern-independent activation of C3 also occurs via hydrolysis, resulting in C3(HO), but the structural details of C3 hydrolysis remain elusive. Plastics are a ubiquitous pollutant, and are rapidly colonized by biofilms that sorb inorganic and organic components, forming the interface between plastics and the environment. This study provides a proof of concept for the use of automated quantitative mineralogy (AQM) to illuminate and analyse the metal and mineral component of the plastisphere on plastics sampled from temperate and tropical aquatic localities. The method is non-destructive and requires minimal sample preparation, providing a 2-dimensional visualisation and semi-quantitative analysis of the arrangement of biogenic and abiogenic components, highlighting potential interactions between these components.

Neurodegenerative diseases are often accompanied by neuroinflammation and impairment of the blood-brain barrier (BBB) mediated by activated glial cells through their release of proinflammatory molecules. To study the effects of glial cells on mouse brain endothelial cells (mBECs), we developed an in vitro BBB model with inflammation by preactivating mixed glial cells (MGCs) with lipopolysaccharide (LPS) before co-culturing with mBECs to study the influence of molecules released by activated MGCs. The response of the mBECs to activated MGCs was compared to direct stimulation with LPS.

Rivers and estuaries are regarded as major pathways of microplastic (MP) transport from terrestrial areas to marine ecosystems. Despite this knowledge on the transport dynamics and fate of MP in freshwater riverine and brackish estuarine waters is limited. Via ex situ settling experiments emulating the Msimbazi River and Estuary in Dar es-Salaam, Tanzania, we demonstrate that flocculation and subsequent settling of positively buoyant MP and fine-grained suspended sediment in riverine and estuarine waters are important for the environmental fate of the plastic particles.

The p90 ribosomal S6 kinases (RSKs) contain two distinct catalytic kinase domains, the N-terminal and C-terminal kinase domains (NTKD and CTKD, respectively). The activation of CTKD is regulated by phosphorylation by extracellular signal-regulated kinase (ERK1/2) and an autoinhibitory αL helix. Through a mutational series in vitro of the RSK CTKDs, we found a complex mechanism lifting autoinhibition that led us to design constitutively active RSK CTKDs.

AbstractMicroplastic particles have become ubiquitous in aquatic environments and can be found in large numbers in riverine, estuarine, and marine settings at the surface of water, in suspension, and as particles deposited at the bed. The transport and settling behavior of small microplastic particles is likely very dependent on interactions with other suspended particles. Here we show from settling tube experiments conducted in the laboratory that fragments and threads of polyvinylchloride microplastic in the size range of 63-125 m readily flocculated with fine-grained natural sediment under relative particle number concentrations that can be observed in nature in high-turbidity estuarine and coastal environments.

Heterozygous mutations in the ATP1A3 gene, coding for an alpha subunit isoform (α3) of Na/K-ATPase, are the primary genetic cause for rapid-onset dystonia-parkinsonism (RDP) and alternating hemiplegia of childhood (AHC). Recently, cerebellar ataxia, areflexia, pes cavus, optic atrophy and sensorineural hearing loss (CAPOS), early infantile epileptic encephalopathy (EIEE), childhood rapid onset ataxia (CROA) and relapsing encephalopathy with rapid onset ataxia (RECA) extend the clinical spectrum of ATP1A3 related disorders. AHC and RDP demonstrate distinct clinical features, with AHC symptoms being generally more severe compared to RDP.

The forcing mechanisms responsible for centennial to millennial variability of mid-latitude storminess are still poorly understood. On decadal scales, the present-day geographic variability of North-Atlantic storminess responds to latitudinal shifts of the North-Atlantic westerly wind-belt under the prime control of the North-Atlantic Oscillation (NAO). An equivalent mechanism operating at centennial to millennial time scales during the Holocene is still to be ascertained, especially owing to the lack of high-resolution and continuous records of past-storminess extending far enough in time.
