Hi-C enables the characterization of the 0conformation of the genome in the three-dimensional nuclear space. This technique has revolutionized our ability to detect interactions between linearly distant genomic sites on a genome-wide scale. Here, we detail a protocol to carry out in situ Hi-C in plants and describe a straightforward bioinformatics pipeline for the analysis of such data, in particular for comparing samples from different organs or conditions.

Article Synopsis
  • Candidemia is frequently caused by a fungal species that has metabolic flexibility, allowing it to utilize various nutrients from the host.
  • Nitrogen assimilation in this fungus involves a mechanism called Nitrogen Catabolite Repression (NCR), regulated by specific GATA factors, particularly highlighting the role of Gln3 in contrast to its function in other species.
  • Recent transcriptional analysis revealed that over 53% of genes were differentially expressed, particularly in amino acid metabolism and ABC transporters, indicating Gln3's complex role in both positive and negative gene regulation, including its surprising impact on fluconazole susceptibility.
