Lysophosphatidylinositol has been previously shown to stimulate cell proliferation in differentiated and in K-ras transformed thyroid cells. Increased levels of lysophosphatidylinositol, but not lysophosphatidylcholine or lysophosphatidylethanolamine, are present in thyroid as well as in other ras-transformed cell lines. We have now investigated the mechanism of action of this lysolipid by analysing its effects in a differentiated thyroid cell line.

The activity of TSH, the main regulator of growth and differentiation in the thyroid, has been mainly related to the activation of the adenylyl cyclase cascade. TSH also activates phospholipase-C and -A2; these effects, however, have been reported to require concentrations of the hormone up to 1000-fold higher than those effective on adenylyl cyclase, suggesting that the main physiological mechanism involved in the action of TSH is the activation of this enzyme. In intact ascites cells magnesium efflux is substantially stimulated by cAMP analogues and by low concentrations of prostaglandins (PGE1 and PGE2) which increases the synthesis of cAMP. cAMP stimulates magnesium efflux by directly stimulating the Na-Mg antiporter rather than determining a magnesium release from mitochondria as reported for other cell types. Protein kinase inhibitors abolish the cAMP stimulated magnesium efflux suggesting that the stimulation is mediated by the phosphorylation of the carrier protein, increasing its affinity for intracellular magnesium.

The aim of these experiments was to study the effect of early enteral nutrition with either standard or enriched (arginine, n-3 fatty acids, RNA) enteral formulas on translocation of bacteria from the gut and acute mortality rate following thermal injury. In the first experiment 60 Balb c mice were gavaged with 10(10)Escherichia coli and received a 20% burn injury. The effectiveness of perioperative administration of thymopentin in preventing postoperative infection was evaluated in 206 patients with cancer (54 gastric, 152 colorectal) who underwent elective major surgery. Comparable subsets of patients were obtained with respect to age (proportion over 65 years) and nutritional status (patients with serum albumin level less than 30 milligrams or weight loss of 10 per cent or more of usual body-weight were considered to be malnourished). Magnesium efflux from intact Ehrlich ascites tumor cells (EATC) has been characterized under 0-trans conditions. It is shown that a magnesium-extruding mechanism operates in these cells which brings about magnesium efflux up to 20% of the cell total content. EATC magnesium efflux is independent from extracellular Ca2+ and its apparent velocity is not affected by [Mg2+]o up to 160 microM.

The efficacy of thymopentin in reducing postoperative infections (PI) was prospectively evaluated in 138 patients with abdominal cancer, who underwent major surgery. Comparable subsets of patients were obtained according to age (cutpoint 65 years) and nutritional status (patients with serum albumin less than 30 g/l or weight loss greater than or equal to 10% with respect to their usual body weight were considered as malnourished). The in vitro effect of increasing concentrations of retinoic acid on the growth of day 7 and day 14 CFU-GM and of pre-CFU-GM from patients with myelodysplastic syndromes has been studied. Unlike in normal cells, where retinoic acid enhances the in vitro growth of myeloid progenitors, there was an overall inhibition of bone marrow progenitors from patients with myelodysplastic syndromes at concentrations of 10(-7) to 10(-6) M retinoic acid, with the exception of day 14 CFU-GM whose growth may be enhanced by 10(-7) M retinoic acid.
