Red cell ferritin content: a re-evaluation of indices for iron deficiency in the anaemia of rheumatoid arthritis.

In iron deficiency anaemia basic red cell content of ferritin is appreciably reduced. This variable was determined in 62 patients with rheumatoid arthritis to evaluate conventional laboratory indices for iron deficiency in the anaemia of rheumatoid arthritis. For 23 patients with rheumatoid arthritis and normocytic anaemia irrespective of plasma ferritin concentration, red cell ferritin content did not differ significantly from that for non-anaemic patients with rheumatoid arthritis.

Comparative in vitro spermicidal effects of (+/-)-gossypol, (+)-gossypol, (-)-gossypol and gossypolone.

The comparative in vitro spermicidal effects of (+)-gossypol, (-)-gossypol and (+/-)-gossypol were evaluated on the spermatozoa of human, monkey, rabbit, mouse, rat and hamster. The spermicidal effects of gossypol isomers were also compared with those of gossypolone, which is a proposed major metabolite of gossypol. Gossypol isomers and gossypolone were all spermicidal.

Plant anticancer agents XXIX. Cleomiscosin A from Simaba multiflora, Soulamea soulameoides, and Matayba arborescens.

Cleomiscosin A (1) has been isolated from Simaba multiflora and Soulamea soulameoides in the Simaroubaceae through bioactivity-directed fractionation and from Matayba arborescens in the Sapindaceae . The structure of cleomiscosin A (1) was established through spectroscopic studies and chemical reactions.

Analysis of gossypol and gossypol-acetic acid by high-performance liquid chromatography.

Gossypol, a bis-sesquiterpenoid cotton pigment, is of current interest as a male fertility-regulating agent. For the purposes of analyzing material to be studied biologically, a method is described for the analysis of gossypol by high-performance liquid chromatography. This has been used for examining the purity of gossypol-acetic acid using a UV-absorbance ratio technique.

Effect of (+)-gossypol on fertility in male hamsters.

(+)-Gossypol was isolated from the bark of Thespesia populnea and tested for its ability to inhibit the fertility of male hamsters. Male hamsters of proven fertility were treated orally for 54 days with 40 mg/kg of (+)-gossypol, 40 mg/kg of racemic gossypol, or 5% gum acacia (vehicle control) and were mated with estrous female hamsters during the fourth and seventh weeks of treatment. Both the control and the (+)-gossypol-treated animals exhibited normal fertility throughout the experiment.

Erythrocyte ferritin content in idiopathic haemochromatosis and alcoholic liver disease with iron overload.

The erythrocyte ferritin content was measured in patients with either idiopathic haemochromatosis or alcoholic liver disease and iron overload to define its value as a marker for an excess of tissue iron. The mean erythrocyte ferritin content in patients with untreated idiopathic haemochromatosis was increased 60-fold and fell with phlebotomy. After phlebotomy many patients had an increased red cell ferritin content despite normal serum ferritin concentrations.

Ibuprofen-induced acute renal failure with acute tubular necrosis.

2 patients with systemic lupus erythematosus and mild renal functional impairment were treated with ibuprofen, one of the phenylproprionic nonsteroidal anti-inflammatory drugs. Within days after the onset of therapy, both developed renal insufficiency manifested by elevated serum creatinine levels, increased proteinuria, and active urinary sediments; 1 patient was oliguric. Renal biopsies disclosed mesangial proliferative lupus glomerulonephritis and acute tubular necrosis, the latter more pronounced in the oliguric patient.

Hormones and factors that stimulate growth of a rat islet tumor cell line in serum-free medium.

A number of hormones and factors were found to stimulate growth of cultured rat islet tumor cells, the RIN-r cell line. A serum-free supplemented medium from RIN-r cells was formulated. It consisted of a 1:1 mixture of Ham's F12 and DME media with the addition of insulin, transferrin, triiodothyronine, prolactin, growth hormone, and an extract of proteose peptone (medium IM).

Antifertility effects of gossypol and its impurities on male hamsters.

Recent papers reporting on the male antifertility effects of gossypol have been criticized due to the possible influence of impurities on efficacy and toxicity. Our laboratory has prepared purified gossypol and a fraction containing the total impurities. The effects of these two substances and impure gossypol on the fertility of male hamsters was evaluated.

Biological and phytochemical investigation of plants XVI: Strumpfia maritima (Rubiaceae).

An aqueous extract of the flowering tops of Strumpfia maritima exhibited antifertility activity in female rats. The extract also contained the flavonol glycoside narcissin. This article represents the first reported isolation of this flavonoid from the Rubiaceae, as well as the first reported phytochemical and pharmacological investigation of the genus Strumpfia.

Hemodynamic assessment of the aortoiliac segment: a prospective study.

A test for assessing stenosis in the aortoiliac segment is described in which femoral pressure is measured directly by continuous monitoring and an intra-arterial injection of papaverine is used to augment blood flow. This test has been described previously but is not widely used. A prospective study of this test was carried out on 64 patients and the results were compared with those obtained by aortography.

Uterotonic effect of Evodia rutaecarpa alkaloids.

Rutaecarpine (1) and dehydroevodiamine (2) were isolated from the unripe fruit of Evodia rutaecarpa. In vitro uterotonic assay on rat uterus showed both alkaloids to be active. Dehydroevodiamine was further shown to be active in the in vivo uterotonic assay using rats as the animal model.

In vitjro spermicidal activity of gossypol.

The in vitro spermicidal effect of gossypol, gossypol acetic acid and gossypol-polyvinylpyrrol idone was investigated. Gossypol-polyvinylmulations. Thus, gossypol in an appropriate chemical form may be useful as a spermicide.

A simple radiometric assay for methotrexate and other folate antagonists.

A rapid radiometric assay for the folate antagonist MTX, based on the inhibition of chicken liver dihydrofolate reductase activity, has been developed. In the system the inhibition by MTX of the reduction of tritiated FH2 is standardized and used to quantitate this antagonist in plasma of subjects receiving high-dose MTX therapy. The assay permits the detection of plasma MTX levels to 5 X 10(-9)M with a coefficient of variation of 15%.

Biological and phytochemical investigation of plants XV. Pteryxia terebinthina var. terebinthina (Umbelliferae).

Aqueous extracts of Pteryxia terebinthina var. terebinthina were shown to possess antiarrhythmic activity. A novel furanocoumarin, pterybinthinone, the known coumarins pteryxin and umbelliferone, and sucrose were isolated from this plant.

Biological and phytochemical investigations of Dianthus barbatus cv. "China Doll" (Caryophyllaceae).

From the aerial parts of Dianthus barbatus cv. "China Doll", two saponins (barbatosides A and B) were isolated and shown to have analgesic and anti-inflammatory activities. The aglycone of each saponin was identified as quillaic acid.

Flavonoid constituents from Eupatorium altissimum L. (Compositae).

An aqueous ethanol extract of Eupatorium altissimum L. (Compositae) showed confirmed activity in the P-388 lymphocytic leukemia assay in mice, and the chloroform solubles showed both cytotoxic activity in the 9KB carcinoma of the nasopharynx cell culture assay and antitumor activity in the P-388 lymphocytic leukemia assay. Two flavones, eupatorin and 5-hydroxy-3',4',6,7-tetramethoxyflavone, were isolated and identified.

Oncogenic and tumor-promoting Spermatophytes and Pteridophytes and their active principles.

A survey and discussion are presented of plants classified as Spermatophyta and Pteridophyta, extracts of which have been shown to be oncogenic or tumor-promoting in animals. The active oncogenic and tumor-promoting principles, where known, have been identified. They represent tannins; pyrrolizidine, indole, tropolone, quinoline, purine, and benzophenanthridine alkaloids; nitroso compounds; triterpene glycosides; lignans; isoflavans; allyl benzenoids; simple (nu-pyrenes; and carbocyclic hydroxy acids.

Isolation of flindersiamine, isoflindersiamine, and a new alkaloid, heliparvifoline, from Helietta parvifolia (Rutaceae).

A phytochemical investigation of the leaves and twigs of Helietta parvifolia (Rutaceae) resulted in the isolation of heliparvifoline, a new furoquinoline alkaloid, in addition to the known bases flindersiamine and isoflindersiamine.
