Artichoke, dandelion, turmeric extracts and rosemary essential oil are commonly used as ingredients in many herbal preparations to treat hepatic and gallbladder disorders. In the present work we compare the activity of each single extract with a commercial mixture for antiproliferative, antiradical and protective effects against induced oxidant stress effect. In ABTS and DPPH tests, turmeric extract is the most active, followed by artichoke and dandelion.

We have investigated the effects of the gastric peptide obestatin injected into the arcuate nucleus of the rat hypothalamus on the hypothalamic mRNA expression of peptides which play master roles as feeding behavior modulators. We have also evaluated the effects of obestatin on dopamine, norepinephrine and serotonin release from rat hypothalamic synaptosomes in vitro. After 4 daily intrahypothalamic injections of obestatin (1 nmol/kg), we recorded a significant reduction of daily caloric intake and body weight gain.

The deleterious effects of cardiac ischemia are ameliorated by ischemic preconditioning (IPC), in which transient ischemia protects against subsequent severe ischemia/reperfusion injury. IPC activates multiple signaling pathways, including the reperfusion injury salvage kinase pathway (mainly PI3K-Akt-glycogen synthase kinase-3beta [GSK3beta] and ERK1/2) and the survivor activating factor enhancement pathway involving activation of the JAK-STAT3 axis.

Novel compounds endowed with a high two-photon absorption (TPA) cross-section and a high singlet oxygen quantum yield, are sought after for their possible application in anti-cancer therapies. In this paper we present a prototype macromolecule bearing a distyrylbenzene dimer as TPA unit and a [60]fullerene moiety for singlet oxygen generation. Two dimers of a heteroaromatic quadrupolar (acceptor-donor-acceptor) chromophore have been synthesized with different interchromophoric distances. Optical spectra of dimers in solution show a red shift of the linear absorption band upon decreasing the interchromophore distance, while fluorescence and two-photon absorption spectra are only marginally affected by the interactions. A bottom up approach is adopted to describe the spectra: via a detailed spectroscopic analysis of the monomeric species in solution, we define an essential-state model for the isolated chromophore and use this information to set up a model for the dimers also accounting for interchromophore electrostatic interactions.

Oxidative stress plays a key role in the pathogenesis of age-related neurodegeneration, and the nonenzymatic production of 8-iso-prostaglandin F(2alpha) (8-iso-PGF(2alpha)) may represent a reliable index of cellular oxidative damage. Garlic (Allium sativum) has been associated with peripheral antioxidant activities and therefore might prevent or reverse 8-iso-PGF(2alpha) production, but scant data are available on its possible neuroprotective effects. Therefore, we have studied the possible antioxidant effects of a garlic extract in rat brain synaptosomes obtained from young (3-month-old) and aged (14-month-old) male Wistar rats that were perfused, in vitro, with graded concentrations of a garlic extract (10-500 microg/mL).

The intensity of the two-photon absorption (TPA) spectrum of a terpyridine ligand acting as a D-pi-A chromophore (D = donor and A = acceptor) is enhanced by a factor of about 2 upon coordination to ZnCl(2). Based on an analysis of linear absorption and fluorescence spectra of both the ligand and its Zn(II) complex, we have defined essential-state models for the two species. Linear and TPA spectra of the ligand are well reproduced in terms of a two-state model accounting for the D-pi-A <--> D(+)-pi-A(-) charge resonance.

Time dependent density functional theory and polarizable continuum model have been used to describe tetra-phenyl-porphyrin derivatives in different solvents and at different pH values. Our results show that last generation functionals are able to describe with good accuracy the photophysics of the title compounds, including different physical-chemical properties of excited electronic states. In particular, the strong modification of polarizabilities induced in some cases by electron excitation could explain a recent hypothesis about NLO characteristics of specific porphyrin adducts.

The aim of the present study is to investigate oxidative stress produced by experimental hypoxia and hyperoxia in young and old pampiniform plexus rats, in order to evaluate the oxidative role of oxygen. Oxidative stress causing molecular and cellular dysfunction increases in hypertension and can therefore be considered a state of oxidative stress. This consideration makes us reflect on the responsibility of oxidative stress in the veins of the pampiniform plexus, notoriously under high hydrostatic pressure.

Glucagon-like peptide 1 (7-36) amide (GLP-1) and exendin-4 are gastrointestinal hormones as well as neuropeptides involved in glucose homeostasis and feeding regulation, both peripherally and at the central nervous system (CNS), acting through the same GLP-1 receptor. Aminergic neurotransmitters play a role in the modulation of feeding in the hypothalamus and we have previously found that peripheral hormones and neuropeptides, which are known to modulate feeding in the central nervous system, are able to modify catecholamine and serotonin release in the hypothalamus. The linear and nonlinear optical properties of the heteroaromatic push-pull-push two-photon absorbing dye N-methyl-2,5-bis[1-(N-methylpyrid-4-yl)ethen-2-yl]-pyrrole ditriflate (PEPEP) are reported. The determination of the two-photon absorption (TPA) cross-section spectrum has been performed with different techniques: femtosecond TPA-white light continuum probe experiments, two-photon-induced fluorescence, and open aperture Z-scan measurements using both nanosecond and femtosecond laser pulses. The measured TPA cross sections and their wavelength dispersion show a marked dependence on the parameters of the laser pulses and on the measurement technique employed.

Singlet oxygen sensitization by organic molecules is a topic of major interest in the development of both efficient photodynamic therapy (PDT) and aerobic oxidations under complete green chemistry conditions. We report on the design, synthesis, biology, and complete spectroscopic characterization (vis-NIR linear and two-photon absorption spectroscopy, singlet oxygen generation efficiencies for both one- and two-photon excitation, electrochemistry, intrinsic dark toxicity, cellular uptake, and subcellular localization) of three classes of innovative singlet oxygen sensitizers pertaining to the family of symmetric squaraine derivatives originating from pi-excessive heterocycles. The main advantage of pi-extended squaraine photosensitizers over the large number of other known photosensitizers is their exceedingly strong two-photon absorption enabling, together with sizable singlet oxygen sensitization capabilities, for their use at the clinical application relevant wavelength of 806 nm.

Cell signalling requires efficient Ca2+ mobilization from intracellular stores through Ca2+ release channels, as well as predicted counter-movement of ions across the sarcoplasmic/endoplasmic reticulum membrane to balance the transient negative potential generated by Ca2+ release. Ca2+ release channels were cloned more than 15 years ago, whereas the molecular identity of putative counter-ion channels remains unknown. In this work we present an investigation of the non-linear optical (NLO) properties of two octupolar chromophores: [Zn(4,4'-bis(dibutylaminostyryl)-[2,2']-bipyridine)(3)](2+) and [Zn(4,4'-bis((E)-2-(N-(TEG)pyrrol-2-yl)vinyl)-[2,2']-bipyridine)(3)](2+) with Zn(ii) as the coordination center, using two-photon emission technique (TPE) in fs-pulse temporal regime. Ca2+ sparks are localized intracellular Ca2+ release events from the sarcoplasmic reticulum in muscle cells that result from synchronized opening of ryanodine receptors (RyR). In mammalian skeletal muscle, RyR1 is the predominant isoform present in adult skeletal fibers, while some RyR3 is expressed during development. Functional studies have revealed a differential role for RyR1 and RyR3 in the overall Ca2+ signaling in skeletal muscle, but the contribution of these two isoforms to Ca2+ sparks in adult mammalian skeletal muscle has not been fully examined.

A strong enhancement of the two-photon absorption (TPA) cross section of tetrakis(4-sulfonatophenyl)porphyrin diacid (H(4)TPPS(2-)) at various wavelengths when a J-type aggregate is formed in water with respect to the one observed for the H(4)TPPS(2-) monomer in a mixture of water, dimethyl sulfoxide (DMSO), and urea is presented. The TPA properties are characterized by the open aperture Z-scan technique and the ultrafast two-photon absorption spectroscopy with white light continuum probe (TPA-WLCP) technique. The microwave-enhanced synthesis, comparative singlet oxygen sensitization efficiency, and nonlinear optical characterization of a new beta-functionalized porphyrin and its copper complex are described. We show that the introduction of a donor-acceptor push-pull conjugated fragment in the beta position strongly perturbs the porphyrin electronic structure leading to a remarkable one- and two-photon NIR absorption enhancement.

This paper presents the synthesis and two photon-induced absorption (TPA) properties of a functionalized distyrylbenzene (DSB) 1 containing a tetra-azacyclododecane (cyclen) receptor for Zn(II). The influence of Zn(II) on one- and two-photon absorption characteristics of 1 has been investigated in dimethyl sulfoxide. The experiments show that the TPA action spectrum of uncomplexed 1, at 750 nm employing nanosecond-long excitation pulses, is 5 times more intense than that of the complexed form.
