-methyl-L-cysteine sulfoxide (SMCO) is a non-protein sulfur-containing amino acid with a variety of functions. There are few reports on the enzymes catalyzing the biosynthesis of SMCO from -methyl-L-cysteine (SMC). In this study, the flavin-containing monooxygenase gene derived from () was heterologously expressed in BL21(DE3) and the enzymatic properties of the expressed protein were analyzed.

Article Synopsis
  • Pediatric lymphoepithelioma-like carcinoma (pLELC) is a rare cancer with uncertain prognosis, and this study focused on its genomic and clinical features among 41 patients from Sun Yat-sen University Cancer Center.
  • The results showed that patients treated with a combination of anti PD-1 therapy and chemotherapy had high progression-free and overall survival rates, with significant immune-related biomarkers identified in the tumor samples.
  • The study suggests that targeting DNA repair pathways could be a viable treatment strategy for pLELC, and advocates for further clinical trials using immune checkpoint inhibitors in this pediatric population.

Background: Immune checkpoint blockade (ICB)-based immunotherapy has inspired new hope for advanced biliary tract cancer (BTC) treatment; however, there are no prior studies that primarily focus on different anatomical types of unresectable BTCs reacting differently to ICB.

Methods: We retrospectively collected data on advanced BTC patients who received anti-programmed cell death protein 1 (anti-PD1) therapy from two affiliated hospitals of Sun Yat-Sen university. The effects of anti-PD1 were compared for different anatomical sites.

Flavin monooxygenases (FMOs) have been widely used in the biosynthesis of natural compounds due to their excellent stereoselectivity, regioselectivity and chemoselectivity. Stenotrophomonas maltophilia flavin monooxygenase (SmFMO) has been reported to catalyze the oxidation of various thiols to corresponding sulfoxides, but its activity is relatively low. Herein, we obtained a mutant SmFMO which showed 4.

Aflatoxins (AFs) are highly carcinogenic metabolites produced by species that can contaminate critical food staples, leading to significant health and economic risks. The cytochrome P450 monooxygenase AflG catalyzes an early step in AF biosynthesis, resulting in the conversion of averantin (AVN) to 5'-hydroxy-averantin. However, the molecular mechanism underlying the AflG-AVN interaction remains unclear.

Purpose: This study aimed to characterize the clinical features, genetic findings, and genotype-phenotype correlations of patients with Leber congenital amaurosis (LCA) or early-onset severe retinal dystrophy (EOSRD) harboring biallelic AIPL1 pathogenic variants.

Design: Retrospective case series.

Methods: This study consecutively enrolled 51 patients from 47 families with a clinical diagnosis of LCA/EOSRD harboring disease-causing variants in the AIPL1 gene, from October 2021 to September 2023.

Bisphenol A (BPA), an endocrine disrupter with estrogen activity, can infiltrate animal and human bodies through the food chain. Enzymatic degradation of BPA holds promise as an environmentally friendly approach while it is limited due to lower stability and recycling challenges. In this study, laccase from Bacillus pumilus TCCC 11568 was expressed in Pichia pastoris (fLAC).

Background: The beneficial effects of first-line programmed death-1 (PD-1) inhibitors plus chemotherapy in patients with low programmed death-ligand 1 (PD-L1)-expressing advanced gastric or gastroesophageal junction (G/GEJ) adenocarcinoma are controversial.

Methods: We conducted a retrospective analysis of patients with G/GEJ adenocarcinoma who had undergone first-line treatment with PD-1 inhibitors plus chemotherapy between October 2017 and May 2022. The primary outcomes were objective response rate (ORR) and progression-free survival (PFS).

Recent advances have revealed that the role of the immune system is prominent in the antitumor response. In the present study, it is aimed to provide an expression profile of tumor-infiltrating lymphocytes (TILs), including mature B cells, plasma cells, and their clinical relevance in neuroblastoma. The expression of CD20 and CD138 was analyzed in the Cangelosi786 dataset (n = 769) as a training dataset and in our cohort (n = 120) as a validation cohort.

Water Distribution Networks (WDNs) are critical infrastructures that ensure a continuous supply of safe water to homes. In the face of challenges, like water scarcity, establishing resilient networks is imperative, especially in regions vulnerable to water crises. This study evaluates the resilience of network designs through graph theory, including its hydraulic feasibility using EPANET software, an aspect often overlooked.

  • A clinical trial was conducted to compare the effectiveness and safety of two treatments—cetuximab plus FOLFOXIRI (triplet therapy) and cetuximab plus FOLFOX (doublet therapy)—in RAS/BRAF wild-type colorectal cancer patients who had unresectable liver metastases.
  • The study enrolled 146 patients across seven medical centers in China between April 2018 and December 2022, assessing their objective response rate and other outcomes such as tumor response and survival.
  • Results showed that the response rates were similar between the two treatment groups (84.7% for triplet vs. 79.7% for doublet), indicating no significant difference in efficacy, and the trial also monitored

Biodegradable microplastics (MPs) are promising alternatives to conventional MPs and are of high global concern. However, their discrepant effects on soil microorganisms and functions are poorly understood. In this study, polyethylene (PE) and polylactic acid (PLA) MPs were selected to investigate the different effects on soil microbiome and C-cycling genes using high-throughput sequencing and real-time quantitative PCR, as well as the morphology and functional group changes of MPs, using scanning electron microscopy and Fourier transform infrared spectroscopy, and the driving factors were identified.

  • - The CAPability-01 trial studied the effectiveness of combining the PD-1 antibody sintilimab with the HDAC inhibitor chidamide, with or without the VEGF antibody bevacizumab, in patients who have advanced colorectal cancer resistant to chemotherapy.
  • - Results showed that the combination therapy (triplet arm) significantly improved progression-free survival and overall response rates compared to the dual therapy (doublet arm), indicating increased treatment efficacy.
  • - Patients experienced various adverse side effects, with two treatment-related deaths reported; however, the analysis suggested the triplet therapy led to enhanced immune activity in the tumor environment.

Reactive oxygen species (ROS) are crucial signaling molecules in plants that play multifarious roles in prompt response to environmental stimuli. Despite the classical thoughts that ROS are toxic when accumulate in excess, recent advances in plant ROS signaling biology reveal that ROS participate in biotic and abiotic stress perception, signal integration, and stress-response network activation, hence contributing to plant defense and stress tolerance. ROS production, scavenging and transport are fine-tuned by plant hormones and stress-response signaling pathways.

Hemicellulose is a highly abundant, ubiquitous, and renewable natural polysaccharide, widely present in agricultural and forestry residues. The enzymatic hydrolysis of hemicellulose has generally been accomplished using β-xylosidases, but concomitantly increasing the stability and activity of these enzymes remains challenging. Here, we rationally engineered a β-xylosidase from to enhance its stability by computation-aided design combining ancestral sequence reconstruction and structural analysis.
