Article Synopsis
  • The study investigates the role of histone demethylase JMJD3 in the nucleus accumbens and its impact on heroin-seeking behavior after a period of abstinence in male rats.
  • Findings show that JMJD3 levels and phosphorylated SMAD1/5 increase following 14 days of heroin abstinence, and manipulating these pathways affects drug-seeking behaviors.
  • The research concludes that JMJD3 is involved in long-term changes linked to heroin relapse, with its effects being regulated by the bone morphogenetic protein (BMP) signaling pathway.

Palmoplantar (PP) psoriasis is considered one of the most hard-to-treat areas with important impact on patients' quality of life, and few data are available about the efficacy of dimethylfumarate (DMF) on these areas. In our noninterventional multicentre retrospective cohort study of patients with PP psoriasis treated with DMF, effectiveness was evaluated as mean pp Psoriasis Area and Severity Index (ppPASI) reduction and as proportion of patients that reach ppPASI 75 at 4, 12, 24 and 48 weeks. The reduction in ppPASI was 23.

Article Synopsis
  • Patients with Alzheimer's disease and their caregivers are at a higher risk for medication-related issues due to aging and the use of multiple medications.
  • A study assessed medication exposure among 73 pairs of Alzheimer's patients and their caregivers, revealing that patients used an average of 6.8 medications, while caregivers used 4.7, with a significant percentage of both groups taking potentially inappropriate medications (60.3% of patients and 47.9% of caregivers).
  • The study highlights the importance of conducting medication reviews not only for patients but also for caregivers, as they are often overlooked in discussions about medication management.

Microsampling, a reduced volume sampling method, has successfully gained attention at the International Conference on Harmonization (ICH) level and established benefits support its use in Toxicokinetic (TK) studies. These improved sampling techniques are less invasive and in large animal species improve animal welfare (refinement). To evaluate if the plasma concentrations of drugs were influenced by the blood sampling method, the traditional method from femoral vein and microsampling from tail vein in Cynomolgus monkeys were compared.

microRNA-22 (miR-22) is a key regulator of lipid and energy homeostasis and represents a promising therapeutic target for NAFLD and obesity. We have previously identified a locked nucleic acid (LNA)-modified antisense oligonucleotide compound complementary to miR-22, designated as RES-010 that mediated robust inhibition of miR-22 function in cultured cells and . In this study we investigated the immune potential of RES-010 in human peripheral blood mononuclear cells (PBMCs).

(1) Background: In their 60-year history, dimethyl fumarate and other salts of fumaric acid have been used for the treatment of psoriasis and other immune-mediated diseases for their immune-modulating properties. Over the years, new mechanisms of action have been discovered for this evergreen drug that remains a first-line treatment for several different inflammatory diseases. Due to its pleiotropic effects, this molecule is still of great interest in varied conditions, not exclusively inflammatory diseases.

We describe here a protocol for the generation of sequence-ready libraries for population epigenomics studies, and the analysis of alignment results. We show that the protocol can be used to monitor chromatin structure changes in populations when exposed to environmental cues. The protocol is a streamlined version of the Assay for transposase accessible chromatin with high-throughput sequencing (ATAC-seq) that provides a positive display of accessible, presumably euchromatic regions.

Background: During the initial assessment of trauma patients, the severity of injury is very often not immediately recognizable. In trauma centers, a total body CT (TBCT) scan is routinely used to evaluate this kind of patients, even if it is burdened with health risk, economical costs, and logistical difficulties.

Aim: We investigated the use of a clinical guide to establish a safe alternative to this routine practice.

Background: Behavioral and psychological symptoms of dementia (BPSD) are frequent and troublesome for patients and caregivers. Considering possible preventive approaches, a better understanding of underlying neural correlates of BPSD is crucial.

Objective: The aim is to assess whether brain regional volume predicts behavioral changes in mild AD.

Background: One of the crucial challenges for the future of therapeutic approaches to Alzheimer's disease (AD) is to target the main pathological processes responsible for disability and dependency. However, a progressive cognitive impairment occurring after the age of 70, the main population affected by dementia, is often related to mixed lesions of neurodegenerative and vascular origins. Whereas young patients are mostly affected by pure lesions, ageing favours the occurrence of co-lesions of AD, cerebrovascular disease (CVD) and Lewy body dementia (LBD).
