Fluorescence sensing is widely used in in vitro detection due to its high sensitivity and rapid result delivery. However, detection systems based on nanomaterials involving complex and cumbersome purification steps can lead to sample loss and significantly reduce the accuracy of the results. To address this issue, we proposed a lanthanide-based aptasensor featuring the target-triggered antenna effect to significantly enhance the time-resolved luminescence (TRL) of chelated Tb combined with a wash-free strategy.

Article Synopsis
  • - Intestinal stem cells (ISCs) play a key role in gut health and recovery from damage, and research showed that Gelsolin KO mice experience more damage and fewer ISCs after radiation exposure.
  • - The study found that the absence of Gelsolin leads to decreased function of ISCs and shuts down the IL-17 signaling pathway, which is crucial for the differentiation of Th17 cells.
  • - Gelsolin supports Th17 cell functionality through the p-STAT3/RORγt pathway, enhancing ISC regeneration and providing protection against various intestinal injuries, not just radiation-induced ones.

Hydrophilic peptides (HPs) play a critical role in the pathogenesis of hepatocellular carcinoma (HCC). However, the comprehensive and in-depth high-throughput analysis of specific changes in HPs associated with HCC remains unrealized, due to the complex nature of biological fluids and the challenges of mining complex patterns in large data sets. The clinical diagnosis of HCC still lacks a non-destructive and accurate classification method, given the limited specificity of widely used biomarkers.

Intestinal stem cells (ISCs) play a crucial role in maintaining the equilibrium and regenerative potential of intestinal tissue, thereby ensuring tissue homeostasis and promoting effective tissue regeneration following injury. It has been proven that targeting Toll-like receptors (TLRs) can help prevent radiation-induced damage to the intestine. In this study, we established an intestinal injury model using IR and evaluated the effects of CL429 on ISC regeneration both in vivo and in vitro.

Ferroptosis, a form of regulated cell death that is driven by iron-dependent phospholipid peroxidation, has been implicated in multiple diseases, including cancer, degenerative disorders and organ ischaemia-reperfusion injury (IRI). Here, using genome-wide CRISPR-Cas9 screening, we identified that the enzymes involved in distal cholesterol biosynthesis have pivotal yet opposing roles in regulating ferroptosis through dictating the level of 7-dehydrocholesterol (7-DHC)-an intermediate metabolite of distal cholesterol biosynthesis that is synthesized by sterol C5-desaturase (SC5D) and metabolized by 7-DHC reductase (DHCR7) for cholesterol synthesis. We found that the pathway components, including MSMO1, CYP51A1, EBP and SC5D, function as potential suppressors of ferroptosis, whereas DHCR7 functions as a pro-ferroptotic gene.

The intricate interplay between the human immune system and cancer development underscores the central role of immunotherapy in cancer treatment. Within this landscape, the innate immune system, a critical sentinel protecting against tumor incursion, is a key player. The cyclic GMP-AMP synthase (cGAS) and stimulator of interferon genes (STING) pathway has been found to be a linchpin of innate immunity: activation of this signaling pathway orchestrates the production of type I interferon (IFN-α/β), thus fostering the maturation, differentiation, and mobilization of immune effectors in the tumor microenvironment.
