This report describes a system of in vivo isolated perfusion of the rat liver. The effects of perfusion with 5-fluorouracil (5-FU) (0.125-1.

We previously reported that administration of a low molecular weight (MW=800) synthetic polymer, NED 137, significantly increases humoral and cellular immune responses in the rat. The effect of NED 137 on the murine humoral response to T-dependent (TD) and T-independent (TI) antigens was studied in C57 BL/6, CBA/J and Balb/c mice. The TD antigens (SRBC, DNP-DA with adjuvant) or TI antigen (DNP-Ficoll) were administered simultaneously with NED 137.

The hypothesis of linkage between HLA and a disease susceptibility (DS) locus (or loci) for type 1 diabetes was tested. HLA segregation was random among 57 non-diabetic sibs but not among 39 diabetic sibs, suggesting that susceptibility to type 1 diabetes may be due to an HLA-linked gene(s). The data did not fit a genetic model involving either a single recessive or dominant gene.

The choice of colloid or crystalloid solutions for fluid resuscitation of critically ill patients remains controversial. Marked reduction of extracellular water is considered an important defect in shock by proponents of crystalloid fluid therapy. Large volumes of crystalloid replenish this extracellular deficit.

A positive crossmatch due to T-cell reactivity against donor cells is considered a strong contraindication to renal transplantation because of the risk of graft loss from rejection. However, the significance of T-cell reactivity before but not at the time of transplantation is unknown. To determine whether transplantation can be successful under these circumstances, graft survival was observed in 15 highly sensitised patients whose T cells were reactive to donor sera before but not at the time of transplantation.

The fidelity of the phase-conjugate signal produced by degenerate four-wave mixing was evaluated by conjugating and recombining the multiple-beam output from a phase grating. A single output beam resulted. If one or more of the beams emerging from the grating were blocked, additional beams appeared whose relative amplitudes agreed with theory for a perfect phase conjugator.

The accessible Ia molecules on the surface of chronic lymphocytic leukemia (CLL) cells were quantitated in the cellular radioimmunoassay with saturating concentrations of monoclonal antibodies. Monoclonal antibody 21w4, like DA/2 antibody, recognizes monomorphic determinants of human Ia antigens. The amount of 21w4 or DA/2 bound to CLL cells derived from eight patients (varying from 2.

This report describes a novel method of immunochemotherapy; the active immunization to the drug 5-fluorouracil (5-FU) with enhanced antitumor activity resulting from its subsequent systemic administration. Two metastasizing carcinomas in the Fischer strain (F344) rat have been used: a chemically induced bladder carcinoma (FBCa) and a spontaneous mammary adenocarcinoma (MACa). Both tumors grow rapidly and result in 100% mortality within 10 wk of implantation.

Human leukemic myeloblasts shed glycoproteins from the cell surface during short-term in vitro culture. Shed surface glycoproteins yield a characteristic profile when studied by gel chromatography, isoelectric focusing, immune precipitation, and polyacrylamide gel electrophoresis. Isolation of immunologically active material yields a compound to approximately 75,000--80,000 daltons, with an isoelectric point of 7.

In order to examine the relevance of the critical body weight hypothesis to menstrual regulation in anorexia nervosa, extensive physical, dietary, and behavioral assessments were made in 40 anorexia nervosa patients immediately before and after treatment, and at 3-month intervals up to 1 year posttreatment. Fifteen patients recovered menses; the body weights of these patients agreed with weights predicted by the critical body weight hypothesis. However, 12 patients with continuing amenorrhea had also achieved similar body weights.

In 31 patients with more than 2 migraine attacks per month the prophylactic effect of tolfenamic acid and placebo was compared in a double-blind crossover study. The patients were treated for 10 weeks with each preparation, with a 2-week break in treatment between the preparations. Tolfenamic acid had a significantly better effect in relation to the preferences of the patients, the number of attacks, the total duration of the attacks, the degree of severity, the number of episodes of vomiting, and work capacity.

The controversial question of an association between human Ia antigens and the FcR for immunoglobulin G has been examined. The experimental basis upon which such a relationship has been predicted was reevaluated. Human allosera with specificity for classical transplantation antigens, HLA-DR, and the related supertypic specificities MB and MT have been tested for their capacities to inhibit homologous EA rosette formation by PBM and B-CLL cells.

The "BB" rat spontaneously develops insulitis followed by impaired glucose tolerance (IGT) and/or an insulin-dependent diabetic syndrome like that in man. All diabetic rats in this study showed marked lymphopenia in blood, lymph nodes, spleen, and thymus. Peripheral blood lymphopenia antedated glucoregulatory disturbances.

An incidence of 9.0 out of 100,000 children under 19 years of age with Type 1 (insulin-dependent) diabetes was detected in Toronto during a 2 year prospective study. An increased number of cases occurred in the winter months of one of the years but not of the other during the study.

Rats exposed to daily 3-hr intermittent food delivery sessions (binging) chronically ingested excessive amounts of either H2O or 5% EtOH (mean 7.1 g/kg/3-hr). Withdrawing the intermittency of the feeding schedule once every 10 days (single-ration probe session) resulted in reduction of fluid intakes (both H2O and EtOH) to the level of animals always given daily single-ration sessions.

This study was undertaken to elucidate the mechanism responsible for the hypolipidemic action of pectin. The experiments reported here were designed to test if direct molecular interactions exist between pectin and lipids. Equilibrium dialysis of pectin and taurocholate showed binding only at high nonphysiological ionic strength.
