Time to viral clearance after successful conservative treatment for high-risk HPV-infected high-grade cervical intraepithelial neoplasia and early invasive squamous cervical carcinoma.

Two-thirds of 152 patients treated for high-grade cervical disease, free of persistence/recurrence, and followed-up both with human papillomavirus (HPV) DNA testing and HPV genotyping cleared their high-risk HPV infection within 1year. Viral clearance continued at diminishing rates during the second and the third year, at the end of which it was virtually complete.

Complex I function in mitochondrial supercomplexes.

This review discusses the functional properties of mitochondrial Complex I originating from its presence in an assembled form as a supercomplex comprising Complex III and Complex IV in stoichiometric ratios. In particular several lines of evidence are presented favouring the concept that electron transfer from Complex I to Complex III is operated by channelling of electrons through Coenzyme Q molecules bound to the supercomplex, in contrast with the hypothesis that the transfer of reducing equivalents from Complex I to Complex III occurs via random diffusion of the Coenzyme Q molecules in the lipid bilayer. Furthermore, another property provided by the supercomplex assembly is the control of generation of reactive oxygen species by Complex I.

Challenging synergistic activity of poplar-bacteria association for the Cd phytostabilization.

The synergistic activity between plants and microorganisms may contribute to the implementation of proactive management strategies in the stabilization of contaminated sites, although heavy metals, such as cadmium (Cd), are potentially toxic to them. The aim of this study was to evaluate the degree of tolerance to Cd contamination (supplying twice 40 mg kg(-1) of Cd) in poplar cuttings [clone I-214, P. × euramericana (Dode) Guinier] inoculated or not with two concentrations of Serratia marcescens strain (1 × 10(7) CFU/g and 2 × 10(7) CFU/g of potting mix).

VIN usual type-from the past to the future.

Usual vulvar intraepithelial neoplasia (uVIN) is the most common VIN type, generally related to a human papillomavirus (HPV) infection, predominantly type 16. The incidence of uVIN has been increasing over the last decades, and a bimodal peak is observed at the age of 40-44 and over 55 years. Almost 40% of patients with uVIN have a past, concomitant or future HPV-associated lesion of the lower genital tract.

Aims: The mitochondrial respiratory chain is recognized today to be arranged in supramolecular assemblies (supercomplexes). Besides conferring a kinetic advantage (substrate channeling) and being required for the assembly and stability of Complex I, indirect considerations support the view that supercomplexes may also prevent excessive formation of reactive oxygen species (ROS) from the respiratory chain. In the present study, we have directly addressed this issue by testing the ROS generation by Complex I in two experimental systems in which the supramolecular organization of the respiratory assemblies is impaired by: (i) treatment either of bovine heart mitochondria or liposome-reconstituted supercomplex I-III with dodecyl maltoside; (ii) reconstitution of Complexes I and III at high phospholipids to protein ratio.

Survival in patients with newly diagnosed conventional glioblastoma: a modified prognostic score based on a single-institution series.

Aims And Background: Recursive partioning analysis (RPA) is commonly used to define the stratification of patients with glioblastoma. Epigenetic silencing of the O6-methylguanine methyltransferase (MGMT) gene by promoter methylation plays an important role in regulating MGMT expression in gliomas and this is an established independent prognostic factor. We tested a prognostic scoring system including all clinical variables used by RPA classification (age, ECOG performance status and type of surgery) and MGMT gene promoter methylation status.

Methods: This retrospective observational cohort study was designed to evaluate ASCUS patients detected through a local cervical cancer screening programme and referred to the Department of Gynaecology and Obstetrics at the S. Orsola-Malpighi University Hospital in Bologna, in the period between January 2000 and December 2007.

Design: The functional study on thyroid tumors with cell oxyphilia (Hürthle cell tumors), characterized by mitochondrial hyperplasia, was carried out in XTC.UC1, and B-CPAP, an oxyphilic and nonoxyphilic thyroid tumor cell line, respectively.

The mitochondrial production of reactive oxygen species in relation to aging and pathology.

Mitochondria are known to be strong producers of reactive oxygen species (ROS) and, at the same time, particularly susceptible to the oxidative damage produced by their action on lipids, proteins, and DNA. In particular, damage to mtDNA induces alterations to the polypeptides encoded by mtDNA in the respiratory complexes, with consequent decrease of electron transfer, leading to further production of ROS and thus establishing a vicious circle of oxidative stress and energetic decline. This deficiency in mitochondrial energetic capacity is considered the cause of aging and age-related degenerative diseases.

The various sources of mitochondrial oxygen radicals: a minireview.

The mitochondrial respiratory chain is the main source of reactive oxygen species in the cell. The main sites of superoxide radical production are within Complexes I and III; however, Complex II, glycerol phosphate dehydrogenase and dihydroorotate dehydrogenase, are also sources of oxygen radicals. By using specific inhibitors within the complexes it is possible to obtain indications at the sites where redox components react with oxygen and in the compartments where they are released.

Diamine oxidase plasma activities after treatment with heparin and jejunal morphometry in untreated coeliac disease.

Diamine oxidase plasma concentrations after treatment with heparin were measured and compared with the surface to volume ratio of jejunal biopsy samples assessed by a morphometric technique in patients with untreated and treated coeliac disease and in biopsied controls. As expected, enzyme activity was significantly lower in patients with untreated coeliac disease than in patients on a gluten-free diet and in biopsied controls. No difference was found between treated patients and biopsied controls.
