Rationale And Objectives: To explore the clinical and computed tomography (CT) characteristics of early-stage lung adenocarcinoma (LADC) that presents with an irregular shape.

Materials And Methods: The CT data of 575 patients with stage IA LADC and 295 with persistent inflammatory lesion (PIL) manifesting as subsolid nodules (SSNs) were analyzed retrospectively. Gasdermin-mediated inflammatory cell death (pyroptosis) can activate protective immunity in immunologically cold tumors. Here, we performed a high-throughput screen for compounds that could activate gasdermin D (GSDMD), which is expressed widely in tumors. We identified 6,7-dichloro-2-methylsulfonyl-3-N-tert-butylaminoquinoxaline (DMB) as a direct and selective GSDMD agonist that activates GSDMD pore formation and pyroptosis without cleaving GSDMD.

The blood proteome holds great promise for precision medicine but poses substantial challenges due to the low abundance of most plasma proteins and the vast dynamic range of the plasma proteome. Here we address these challenges with NUcleic acid Linked Immuno-Sandwich Assay (NULISA™), which improves the sensitivity of traditional proximity ligation assays by ~10,000-fold to attomolar level, by suppressing assay background via a dual capture and release mechanism built into oligonucleotide-conjugated antibodies. Highly multiplexed quantification of both low- and high-abundance proteins spanning a wide dynamic range is achieved by attenuating signals from abundant targets with unconjugated antibodies and next-generation sequencing of barcoded reporter DNA.

Background And Objectives: White matter hyperintensities (WMH) correlate with Alzheimer disease (AD) biomarkers cross-sectionally and modulate AD pathogenesis. Longitudinal changes have been reported for AD biomarkers, including concentrations of CSF β-amyloid (Aβ) 42, Aβ40, total tau and phosphorylated tau181, standardized uptake value ratio from the molecular imaging of cerebral fibrillar Aβ with PET using [C] Pittsburgh Compound-B, MRI-based hippocampal volume, and cortical thickness. Correlations between established AD biomarkers and the longitudinal change for WMH have not been fully evaluated, especially among cognitively normal individuals across the adult life span.

For the purpose of obtaining red-light phosphors with excellent luminescence thermal stability, a series of GdAlO:Eu (GAO:Eu) phosphors were synthesized by combining the sol-gel method with high-temperature calcination, and a detailed series of study and analysis of their room temperature and high temperature luminescence properties was carried out. In GAO, the emission peaks corresponding to the D → F ( = 0, 1, 2, 3 and 4) transitions of Eu were observed at 578, 590, 610, 654, and 707 nm, with the strongest emission peak at 610 nm, and the obtained samples were red-light phosphors. The sample GAO:Eu synthesized by combining the sol-gel method with high-temperature calcination has a negative thermal quenching (NTQ) effect, and the best doped sample GAO:0.

Angiotensin-converting enzyme 2 (ACE2) and accessory proteases (TMPRSS2 and CTSL) are needed for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) cellular entry, and their expression may shed light on viral tropism and impact across the body. We assessed the cell-type-specific expression of ACE2, TMPRSS2 and CTSL across 107 single-cell RNA-sequencing studies from different tissues. ACE2, TMPRSS2 and CTSL are coexpressed in specific subsets of respiratory epithelial cells in the nasal passages, airways and alveoli, and in cells from other organs associated with coronavirus disease 2019 (COVID-19) transmission or pathology.
