Osteoporosis is a frequent complication of chronic inflammatory diseases and increases in the pro-inflammatory cytokines make an important contribution to bone loss by promoting bone resorption and impairing bone formation. Omentin-1 is a newly identified adipocytokine that has anti-inflammatory effects, but little is known about the role of omentin-1 in inflammatory osteoporosis. Here we generated global knockout () mice and demonstrated that depletion of omentin-1 induces inflammatory bone loss-like phenotypes in mice, as defined by abnormally elevated pro-inflammatory cytokines, increased osteoclast formation and bone tissue destruction, as well as impaired osteogenic activities.

Femoral neck geometric parameters (FNGPs) are closely related to the strength of the femoral neck and the risk of fragility fractures. No reference database is available for FNGPs for Chinese population, and gender-related differences in FNGPs as well as their association with the risk of femoral neck fractures are unknown. This investigation aimed to set up reference databases for FNGPs, understand gender-related differences in FNGPs, and examine the association between FNGPs and the risk of osteoporotic fractures of the femoral neck.

Article Synopsis
  • * The study shows that vaspin reduces the osteogenic differentiation of preosteoblast cells in a way that depends on its concentration, largely by increasing levels of miRNA-34c, which then inhibits Runx2, a key gene for bone formation.
  • * Vaspin activates the PI3K-Akt signaling pathway, and when this pathway is blocked, the inhibitory effects of vaspin on osteogenic differentiation are lessened; a feedback loop between PI3K-Akt and miR-34c influences their activities in regulating

Bone regeneration is a promising avenue for osteoporosis (OP) treatment. It consists of several procedures, such as the osteogenic differentiation of mesenchymal stem cells as well as the proliferation and maturation of osteoblasts. Many epigenetic mechanisms are involved in the modulation of bone regeneration, including DNA methylation, histone modifications, and microRNA regulation.

We previously reported that Runx2/miR-3960/miR-2861 regulatory feedback loop stimulates osteoblast differentiation. However, the effect of this feedback loop on the osteogenic transdifferentiation of vascular smooth muscle cells (VSMCs) remains unclear. Our recent study showed that miR-2861 and miR-3960 expression increases significantly during β-glycerophosphate-induced osteogenic transdifferentiation of VSMCs.

Article Synopsis
  • Arterial calcification is common and linked to higher cardiovascular death rates, particularly in patients with end-stage renal disease (ESRD) or diabetes.
  • Recent studies suggest that various genetic and environmental factors contribute to this condition, but many uncertainties remain.
  • Epigenetics, which deals with changes in gene regulation without altering the DNA sequence, is emerging as a key area of research that may help understand arterial calcification better and inform potential therapeutic strategies.

Bone marrow mesenchymal stem cells (BMSCs) exhibit an age-dependent reduction in osteogenesis that is accompanied by an increased propensity toward adipocyte differentiation. This switch increases adipocyte numbers and decreases the number of osteoblasts, contributing to age-related bone loss. Here, we found that the level of microRNA-188 (miR-188) is markedly higher in BMSCs from aged compared with young mice and humans.

Article Synopsis
  • The study aimed to explore how variations in the LRP5 gene affect the effectiveness of alendronate treatment in women with osteoporosis in China.
  • The research involved 639 postmenopausal women who were treated with alendronate, and the A1330V polymorphism in LRP5 was examined alongside changes in bone mineral density and bone turnover markers before and after treatment.
  • Results showed that women with CC and CT genotypes experienced more significant improvements in lumbar spine bone density and decreases in specific bone turnover markers than those with the TT genotype, indicating that the TT genotype may signal a weaker response to the medication.

Arterial calcification is a complex and active regulated process, which results from a process of osteoblastic differentiation of vascular smooth muscle cells (VSMCs). Leptin, the product of the ob gene, mainly regulates food intake and energy expenditure and recently has been considered to be correlated with the arterial calcification. However, the mechanisms of the effects of leptin on osteoblastic differentiation of VSMCs are unknown.

Ghrelin is a 28-amino-acid peptide that acts as a natural endogenous ligand of the growth hormone secretagogue receptor (GHSR) and strongly stimulates the release of growth hormone from the hypothalamus-pituitary axis. Previous studies have identified the important physiological effects of ghrelin on bone metabolism, such as regulating proliferation and differentiation of osteoblasts, independent of GH/IGF-1 axis. However, research on effects and mechanisms of ghrelin on osteoblast apoptosis is still rare.

It has been presumed that adipokines deriving from adipose tissue may play important roles in bone metabolism. Omentin-1, a novel adipokine, which is selectively expressed in visceral adipose tissue, has been reported to stimulate proliferation and inhibit differentiation of mouse osteoblast. However, little information refers to the effect of omentin-1 on human osteoblast (hOB) proliferation.

Wisp3 gene mutation was shown to cause spondyloepiphyseal dysplasia tarda with progressive arthropathy (SRDT-PA), but the underlying mechanism is not clear. To clarify this mechanism, we constructed the wild and mutated Wisp3 expression vectors and transfected into human chondrocytes lines C-20/A4; Wisp3 proteins subcellular localization, cell proliferation, cell apoptosis, and Wisp3-mediated gene expression were determined, and dynamic secretion of collagen in transfected chondrocytes was analyzed by (14)C-proline incorporation experiment. Mutated Wisp3 protein increased proliferation activity, decreased apoptosis of C-20/A4 cells, and aggregated abnormally in cytoplasm.
