Efficacy and safety of nonsteroidal antiinflammatory drugs for cancer pain: a meta-analysis.

Purpose: To assess the efficacy and safety of nonsteroidal antiinflammatory drugs (NSAIDs) in the treatment of cancer pain by meta-analyses of the published randomized control trials (RCTs).

Patients And Methods: Twenty-five studies met inclusion criteria for analysis. Of these, 13 tested a single-dose effect, nine multiple-dose effects, and three both single- and multiple-dose effects of 16 different NSAIDs in a total of 1,545 patients.

ATPase activity associated with the uncoating of clathrin baskets by Hsp70.

In the presence of ATP, bovine brain hsp70 has been shown to remove clathrin from bovine brain clathrin-coated vesicles in a rapid stoichiometric initial burst followed by slow steady-state uncoating. In addition, it has been found recently that a 100-kDa cofactor is required for hsp70 to uncoat clathrin baskets prepared with the assembly protein AP-2. In this study the ATPase activity associated with uncoating was investigated, with baskets formed from clathrin and assembly proteins.

The effects of the clinically tested NMDA receptor antagonist memantine on carrageenan-induced thermal hyperalgesia in rats.

This study tested the prophylactic and the therapeutic efficacy of memantine (1-amino-3,5-dimethyl-amandate), a clinically tested N-methyl-D-aspartate (NMDA) receptor antagonist, in suppressing carrageenan-induced thermal hyperalgesia in rats. Complex formation between clathrin and uncoating ATPase.

The bovine brain uncoating ATPase, a constitutive 70-kDa heat shock protein, uncoats clathrin-coated vesicles in an ATP-dependent reaction. The uncoating ATPase-clathrin complex formed from the uncoating reaction was compared to the complex formed by directly binding free clathrin to uncoating ATPase. The amount of the latter complex shows a simple hyperbolic dependence on either free clathrin or free uncoating ATPase concentration, whichever is in excess, with a binding stoichiometry of one uncoating ATPase per clathrin heavy chain.

Characterization of nucleotide-free uncoating ATPase and its binding to ATP, ADP, and ATP analogues.

The interactions of the 70-kDa heat-shock proteins (hsp70s) with their protein substrates appear to be regulated by bound nucleotide. Previous work has shown that the nucleotide binding site of the bovine brain uncoating ATPase, a constitutive member of the hsp70 family, crystallographically resembles the nucleotide binding site of actin and, like actin, the uncoating ATPase has a strongly bound ADP which cannot be removed by dialysis or treatment with ethylenediaminetetraacetic acid (EDTA). This suggests that, like the bound nucleotide of actin, it may be required for the enzyme to retain its native structure.

Smoking, gender, and age as risk factors for site-specific intraoral squamous cell carcinoma. A case-series analysis.

Background: Studies assessing risk factors for oral cancer do not generally report results for specific oral sites. The purpose of the current study was to examine differences in the distribution of age, gender, and tobacco use by intraoral site in a series of oral cancer cases.

Methods: Information on gender, age at diagnosis, and lesion location was obtained for all incident cases of oral squamous cell carcinoma diagnosed through the University of Connecticut Oral Pathology Biopsy Service during the period 1987-1991 (N = 150).

Determination of etoposide in blood by liquid chromatography with electrochemical detection.

A liquid chromatographic (LC) method for determination of etoposide in dog blood is described. The technique includes solvent extraction of etoposide using a dichloroethane-hexane mixture and reconstitution of the drug in an aqueous reconstitution solution. The samples are analyzed by reversed-phase LC with electrochemical detection.

Immediately after clathrin-coated pits pinch off from the cell membrane to form clathrin-coated vesicles, clathrin dissociates from the vesicles. In vitro studies suggest that this dissociation is carried out by the uncoating ATPase, a constitutive member of the 70-kDa heat shock family. Aside from the requirement for ATP, nothing is known about the regulation of the uncoating process.

Laparoscopic ablation of endometriosis using the cavitational ultrasonic surgical aspirator.

Surgical modalities such as electrosurgery and lasers have been used for many years to treat endometriosis. They are relatively unselective with wide scatter, however, leading to the potential for significant tissue damage and injury. As an alternative, a technique for performing laparoscopic excision and adhesiolysis using a cavitational ultrasonic surgical aspirator (CUSA) was developed and studied in 15 patients.

A method for estimation of extracellular concentration of compounds by microdialysis using urea as an endogenous recovery marker in vitro validation.

A new, practical method called microdialysis has been developed for the estimation of free, unbound drug concentrations in the interstitial fluid. This method is based on the use of urea as an endogenous recovery marker. Urea freely distributes throughout the body water compartment, and its plasma concentration can be used as an accurate measure of its interstitial levels.

The NMDA antagonist Memantine blocks pain behavior in a rat model of formalin-induced facial pain.

Recent studies have provided evidence that excitatory amino acid antagonists can exert analgesic effects in animals. These studies, however, have focused primarily on phasic pain or hyperalgesia rather than tonic pain. The present study evaluates the effects of systemic administration of Memantine (1-amino-3,5-dimethyl-adamantane), a clinically used N-methyl-D-aspartate (NMDA) receptor antagonist, on formalin-induced phasic and tonic pain behavior in the rat.

Nucleotide binding properties of bovine brain uncoating ATPase.

Many functions of the 70-kDa heat-shock proteins (hsp70s) appear to be regulated by bound nucleotide. In this study we examined the nucleotide binding properties of purified bovine brain uncoating ATPase, one of the constitutively expressed members of the hsp70 family. We found that uncoating ATPase purified by ATP-agarose column chromatography retained one ADP molecule bound per enzyme molecule which could not be removed by extensive dialysis.

Pharmacokinetic measurement of drugs in lung epithelial lining fluid by microdialysis: aminoglycoside antibiotics in rat bronchi.

A novel technique for in vivo intrabronchial pharmacokinetic measurements using microdialysis is described. The technique was applied to the measurement of tobramycin and gentamicin in the anesthetized rat. The concentration versus time profiles of the drugs in the lung epithelial lining fluid (ELF) following intravenous bolus administration were determined.

Incidental oral hairy leukoplakia in immunocompetent persons. A report of two cases.

In this brief article we report on two HIV-negative patients with documented oral hairy leukoplakia who had no known risk factors for HIV infection nor any evidence of other forms of immune suppression. Therefore we conclude that in some instances hairy leukoplakia can represent an isolated and innocuous Epstein-Barr virus infection.

Lichenoid lesions of oral mucosa. Diagnostic criteria and their importance in the alleged relationship to oral cancer.

Lichen planus and other lichenoid disorders of oral mucosa occur commonly, and yet they are poorly understood. Further, the question of the premalignant potential of oral lichen planus remains deeply mired in controversy. The object of this article is to delineate these problems, clarify the issues, and present evidence to support the position that true lichen planus of oral mucosa is not only less common than generally appreciated but also has no inherent predisposition to become malignant.

Lichen planus and oral cancer: is there a connection between the two?

Objective: To evaluate the effect of gonadotropin-releasing hormone agonist (GnRH-a) when given through stimulation with gonadotropins versus stopping it as stimulation was initiated versus a control group unprepared in this manner.
