The trial of experimental vaccine consisting of protective protein antigens of P. aeruginosa cell wall was carried out on 114 volunteers. The vaccine proved to be faintly reactogenic and induced the formation of specific humoral immunity in 98% of the volunteers who retained a high level of anti-P.

A preclinical study of seven batches of lactoglobulin, a new biological preparation against opportunistic bacteria and salmonellae, has been carried out. High antibacterial activity of the preparation with respect to the virulent forms of Salmonella typhimurium, Salmonella enteritidis, Pseudomonas aeruginosa, Klebsiella pneumoniae, Proteus vulgaris, Proteus mirabilis has been established. The preparation has been shown to be safe and nontoxic.

Synthetic polysaccharide (S-PS) containing aglycone-spacer with a free amino group was really alpha 1,6-mannan with Cn approximately 10. S-PS was transformed into isothiocyanate derivative by treating it with thiophosgene and engaged into reaction with amino group of bovine serum albumin (BSA) lysine residues. Partially purified water extract was obtained from the initial water extract by ultracentrifugation. Nine protein fractions differing in molecular weight, homogeneity and the content of lipopolysaccharide (LPS) were obtained by stepwise precipitation with ammonium sulfate and subsequent fractionation in columns packed with Sephadex G-100 and DEAE cellulose. Two protein fractions with a molecular weight of 30000 and 40000 daltons were practically free of LPS.

To determine the content of lipopolysaccharide (LPS) in P. aeruginosa aqueous extracts the ELISA was used. Membrane filtration and ultracentrifugation followed by precipitation with ammonium sulfate at 80% saturation and gel filtration on Sephadex G-100 have proved to be the most effective methods for purification of the aqueous extract from LPS.

The preliminary study of the safety, reactogenicity and immunological effectiveness of polyvalent vaccine prepared from Pseudomonas aeruginosa protective antigens was carried out. A total of 191 batches of commercial human gamma globulin were studied with the aim of detecting natural antibodies by means of cross and countercurrent electrophoresis, immunodiffusion in agar gel and the passive hemagglutination test. Commercial human gamma globulin preparations were found to contain, to a greater or lesser extent, natural antibodies to the cell-wall O-antigen of Ps. aeruginosa and the antigens of its extracellular slime (capsule); the titers of these antibodies were 1 : 64 to 1 : 256 in 75-85% of cases, 1 : 512 and greater in 10-11% of cases.
