Hodgkin Reed-Sternberg (HRS) cells of classic Hodgkin lymphoma (cHL), like many solid tumors, elicit ineffective immune responses. However, patients with cHL are highly responsive to PD-1 blockade, which largely depends on HRS cell-specific retention of MHC class II and implicates CD4 T cells and additional MHC class I-independent immune effectors. Here, we utilize single-cell RNA sequencing and spatial analysis to define shared circulating and microenvironmental features of the immune response to PD-1 blockade in cHL.

Several therapeutic agents have been approved for treating multiple myeloma (MM), a cancer of bone marrow resident plasma cells. Predictive biomarkers for drug response could help guide clinical strategies to optimize outcomes. Here, we present an integrated functional genomic analysis of tumor samples from MM patients that were assessed for their ex vivo drug sensitivity to 37 drugs, clinical variables, cytogenetics, mutational profiles, and transcriptomes.

Importance: Aggressive thyroid carcinoma, including radioiodine refractory (RAIR) differentiated thyroid carcinoma (DTC), medullary thyroid carcinoma (MTC), and anaplastic thyroid carcinoma (ATC), are associated with significant morbidity and mortality and have limited therapeutic options. Distinct immune profiles have been identified in thyroid cancer subtypes suggesting they may be susceptible to immune checkpoint inhibition.

Objective: To evaluate the efficacy of anti-programmed cell death 1 nivolumab and anti-cytotoxic lymphocyte-associated protein 4 ipilimumab in patients with aggressive thyroid carcinoma.

Article Synopsis
  • The study investigates the response to first-line PD-1 inhibitors, pembrolizumab and cemiplimab, in patients with metastatic NSCLC, focusing on those with different PD-L1 tumor proportion scores (TPS).
  • It compares survival rates between patients with a PD-L1 TPS of 90% or higher and those with a score of 50% to 89%, finding significantly better outcomes for the higher TPS group in both treatment cohorts.
  • Genomic profiling identified specific mutations more prevalent in lower PD-L1 expression tumors, highlighting key biological differences that may influence treatment responses.

Article Synopsis
  • - The study explores the combination of pembrolizumab (an anti-PD1 therapy) and trebananib (an angiopoietin inhibitor) in patients with metastatic ovarian cancer and microsatellite stable (MSS) colorectal cancer, as both cancers show resistance to PD1 immunotherapy.
  • - Results indicate that the highest tolerated dose of the combination therapy is trebananib at 30 mg/kg weekly plus pembrolizumab at 200 mg every 3 weeks, with a modest overall response rate of 7.3%, including durable responses in three MSS CRC patients.
  • - The successful patients exhibited particular tumor characteristics, such as left-sided CRC and no liver metastases; highlighting the need for further research into how
