Background: Crizanlizumab is a novel inhibitor of P-selectin, a key player in multicellular adhesion and inflammatory signaling, that leads to vaso-occlusion in sickle cell disease (SCD).

Objectives: The SOLACE-adults study evaluated the pharmacokinetics, pharmacodynamics (P-selectin inhibition), safety, and efficacy of crizanlizumab, with or without hydroxyurea/hydroxycarbamide, in patients with SCD.

Design: Phase II, single-arm, multicenter study.

Backgruound: Social determinants of health (SDOH) have been associated with diabetes risk; however, their association with cardiovascular disease (CVD) events in individuals with diabetes is poorly described. We hypothesized that a greater number of SDOH among individuals with diabetes would be associated with a higher risk of CVD events.

Methods: The REasons for Geographic and Racial Differences in Stroke (REGARDS) study is a national, biracial cohort of 30,239 individuals ≥45 years old recruited in 2003-2007.

Background: The coronavirus disease 2019 (COVID-19) pandemic was characterized by rapid evolution of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants, affecting viral transmissibility, virulence, and response to vaccines/therapeutics. EMPATHY (NCT04828161), a phase 2 study, investigated the safety/efficacy of ensovibep, a multispecific designed ankyrin repeat protein (DARPin) with multivariant in vitro activity, in ambulatory patients with mild to moderate COVID-19.

Methods: Nonhospitalized, symptomatic patients (N = 407) with COVID-19 were randomized to receive single-dose intravenous ensovibep (75, 225, or 600 mg) or placebo and followed until day 91.

Article Synopsis
  • There’s no standardized method to screen for social risk factors in adults, so this study aimed to create mortality risk prediction models using social determinants of health (SDoH) for better clinical risk assessment.
  • The research utilized data from a national cohort (REGARDS) of over 20,000 adults aged 45 and older to analyze how individual, area-level, and business-level SDoH relate to all-cause mortality.
  • The findings indicated that while individual-level SDoH improved mortality predictions compared to basic demographic data, adding area and business-level SDoH provided minimal benefits, highlighting the importance of SDoH in identifying high-risk individuals for further intervention.

The V-O-N coating set was produced at different relative oxygen concentrations of O(x) = O/(N +O) using cathodic arc evaporation. The aim of the research was to determine the effect of oxygen on coating properties. The coatings' composition and structural properties (X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX)) and mechanical properties-hardness, adhesion, and wear resistance (nano-indenter, scratch tester, ball-on-disc tester)-were extensively investigated.
