In this study, we isolated a new isoflavanostilbene maackiapicevestitol () as a mixture of two stable conformers and as well as five previously known dimeric and monomeric stilbens: piceatannol (), maackin (), scirpusin A (), maackiasine (), and maackolin () from heartwood, using column chromatography on polyamide, silicagel, and C-18. The structures of these compounds were elucidated by NMR, HR-MS, and CD techniques. Maksar obtained from heartwood and polyphenolics - possessed moderate anti-HSV-1 activity in cytopathic effect (CPE) inhibition and RT-PCR assays.
View Article and Find Full Text PDFAdjuvant activity of fucoidans in experimental vaccine compositions with ovalbumin was studied on a mouse model. Compositions with sulfated polysaccharides from brown alga Fucus evanescens (native fucoidan in combination with polyphenols, and a product of fucoidan enzymatic hydrolysis) induced multiple productions of antigen-specific antibodies - total IgG, its isotypes IgG1 and, especially, IgG2a, in comparison with an individual ovalbumin. The adjuvant effect of native and structurally modified fucoidans is slightly inferior to that of the traditional licensed aluminum hydroxide adjuvant.
View Article and Find Full Text PDFThе study presents the results of a comparative evaluation of the effect of structural modifications of fucoidans from the brown alga Fucus evanescens (native, highly purified рroduct of fucoidan enzymatic hydrolysis, a new regular 1→3;1→4-α-L-fucan, sulphated mainly at C2 and acetylated at C4 of the fucose residue) on the effector functions of innate and adaptive immunity cells in vitro and in vivo. Using flow cytometry, we found that all examined fucoidans induce the maturation of dendritic cells, enhance the ability of neutrophils to migrate and adhere, activate monocytes and enhance their antigen-presenting functions, and increase the cytotoxic potential of natural killers. Fucoidans increase the production of hepatitis B virus (HBs) specific IgG and cytokine Th1 (IFN-γ, TNF-α) and Th2 (IL-4) profiles in vivo.
View Article and Find Full Text PDFThe effect of sulfated polysaccharide from brown alga Fucus evanescens (fucoidan) administered via different routes (peroral and parenteral) on the dynamic of some lipid metabolism parameters and markers of systemic inflammation in mice with experimental dyslipidemia induced by prolonged administration of poloxamer P-407. It was found that fucoidan corrected the main parameters of lipid metabolism, reduced the level of endothelial dysfunction marker endothelin-1 and proinflammatory cytokines TNFα, IFNγ in blood serum in animals with experimental dyslipidemia. These findings open prospects for using fucoidan in the complex treatment of metabolic disorders and atherosclerotic inflammation.
