The formation constants of CaUO(CO) complexes were determined directly using capillary electrophoresis coupled with inductively coupled plasma mass spectrometry (CE-ICP-MS) in 0.1 M NaCl and at room temperature. Instead of conventional fused silica, polyetheretherketone (PEEK), a neutral organic material, was used as the capillary material to avoid interactions between uranyl and silica.

The objective of this work was to develop an actinide-specific monolithic support in capillary designed to immobilize precise Pu:Am ratios and its coupling to inductively coupled plasma mass spectrometry (ICP-MS) for immobilized metal affinity chromatography applications. This format offers many advantages, such as reducing the sample amount and waste production, which are of prime importance when dealing with highly active radioelements. Four organic phosphorylated-based monoliths were synthesized in situ through UV photo-polymerization in capillary and characterized.

JRC-Geel organised a Europe-wide proficiency test on gross alpha/beta activity concentration measurements in water with 154 participants from the field of environmental radioactivity monitoring. The performance of the participants was evaluated by comparing their results to the reference value using percentage deviation, z-score and zeta-score. Many of the participants' results deviated several orders of magnitude from the reference values regardless of the techniques used suggesting that the radioactivity monitoring of drinking water using gross-counting in Europe needs improving.

The Hippo pathway is an evolutionarily conserved kinase cascade involved in the control of tissue homeostasis, cellular differentiation, proliferation, and organ size, and is regulated by cell-cell contact, apical cell polarity, and mechanical signals. Miss-regulation of this pathway can lead to cancer. The Hippo pathway acts through the inhibition of the transcriptional coactivators YAP and TAZ through phosphorylation.

We report the design and evaluation of pH responsive luminescent europium(iii) probes that allow conjugation to targeting vectors to monitor receptor internalisation in cells. The approach adopted here can be used to tag proteins selectively and to monitor uptake into more acidic organelles, thereby enhancing the performance of time-resolved internalisation assays that require pH monitoring in real time.

Capillary electrophoresis (CE) was hyphenated to multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS) to determine the model age of a highly enriched uranium (HEU) sample using the U/Th radiochronometer. The use of hydroxymethylbutyric acid (HMBA) as the CE electrolyte was investigated, and a complexation stacking method was developed to increase the thorium signal obtained. The age of the material was determined by measuring the Th content of the HEU sample using isotope dilution in conjunction with the CE-MC-ICP-MS protocol.

Although G protein-coupled receptor (GPCR) oligomerization is a matter of debate, it has been shown that the nature of the GPCR partners within the oligomers can influence the pharmacological properties of the receptors. Therefore, finding specific ligands for homo- or hetero-oligomers opens new perspectives for drug discovery. However, no efficient experimental strategy to screen for such ligands existed yet.

Background: High-yielding dairy cows are prone to oxidative stress due to the high metabolic needs of homeostasis and milk production. Oxidative stress and inflammation are tightly linked; therefore, anti-inflammatory and/or natural antioxidant compounds may help improve mammary cell health. Baicalin, one of the major flavonoids in , has natural antioxidant and anti-inflammatory properties in various cell types, but its effects on bovine mammary epithelial cells (BMECs) have not been investigated.

Systemic inflammation is common in early lactation dairy cows and is associated with decreased milk production. The Scutellaria baicalensis plant contains flavonoids with anti-inflammatory and anti-oxidative properties, which may counteract the inflammatory state in early lactation dairy cows. The objective of this experiment was to determine whether Scutellaria baicalensis extract (SBE), a source of bioactive flavonoids, would alter the adaptation to lactation.

The YAP protein is a co-transcription factor increasing the expression of genes involved in cell proliferation and repressing the expression of genes important for cell differentiation and apoptosis. It is regulated by several inputs, like the Hippo pathway, through the action of kinases that phosphorylate YAP on several residues. The level of phosphorylation of the residues serine 127 (S127) of YAP is generally assessed in cellular models, native tissues, and organs, as a marker of YAP activity and location, and is regulated by numerous partners.

Article Synopsis
  • Group-III metabotropic glutamate (mGlu) receptors are key in regulating synapses and are potential targets for treating conditions like Parkinson's, autism, and pain.
  • Potential drug options include orthosteric agonists, which have limitations such as low specificity and difficulty crossing the blood-brain barrier, and allosteric modulators, which offer more specificity but have issues with bioavailability.
  • This study used a Tag-lite assay to analyze the effects of several agonists and allosteric modulators on various G protein-coupled receptors, finding that while orthosteric agonists showed no off-target activity, some allosteric modulators did have effects on non-target receptors.

Microglia, the tissue-resident macrophages of the CNS, represent major targets for therapeutic intervention in a wide variety of neurological disorders. Efficient reprogramming protocols to generate microglia-like cells in vitro using patient-derived induced pluripotent stem cells will, however, require a precise understanding of the cellular and molecular events that instruct microglial cell fates. This remains a challenge since the developmental origin of microglia during embryogenesis is controversial.

Precise isotopic and elemental characterization of spent nuclear fuel is a major concern for the validation of the neutronic calculation codes and waste management strategy in the nuclear industry. Generally, the elements of interest, particularly U and Pu which are the two major elements present in spent fuel, are purified by ion exchange or extractant resins before off-line measurements by thermal ionization mass spectrometry. The aim of the present work was to develop a new analytical approach based on capillary electrophoresis (CE) hyphenated to a multicollector inductively coupled plasma mass spectrometer (MC-ICPMS) for online isotope ratio measurements.
