Objective: Low incidence of Pott's Puffy tumor (PPT) has caused studying risk factors and recurrences of the disease to be difficult. We used the comparatively increased incidence at our institution to evaluate potential risk factors for the disease process itself and prognostic factors for recurrence of the disease.

Methods: Single institutional retrospective chart review identified 31 patients from 2010 to 2022 with PPT compared with a control group of 20 patients with either chronic rhinosinusitis or recurrent sinusitis.

Article Synopsis
  • The UK National Institute for Health and Care Excellence (NICE) reviewed pegcetacoplan by Apellis Pharmaceuticals/Sobi as a treatment for adults with uncontrolled anemia from paroxysmal nocturnal hemoglobinuria (PNH), comparing it to eculizumab and ravulizumab.
  • The Liverpool Reviews and Implementation Group acted as the Evidence Review Group and aimed for a Fast Track Appraisal due to the company's low cost-effectiveness ratio goal.
  • Clinical evidence showed pegcetacoplan significantly improved hemoglobin levels compared to eculizumab, but indirect comparisons with ravulizumab were deemed unreliable by both the company and the ERG.

Sirtuins (SIRTs) are a class of nicotinamide adenine dinucleotide (NAD)-dependent protein histone deacetylases (HDACs) that are evolutionarily conserved from bacteria to mammals. This group of enzymes catalyses the reversible deacetylation of lysine residues in the histones or non-histone substrates using NAD as a cosubstrate. Numerous studies have demonstrated that the aberrant enzymatic activity of SIRTs has been linked to various diseases like diabetes, cancer, and neurodegenerative disorders.
