In connection with the introduction of economic sanctions against the Russian Federation by the United States, the European Union, Japan, and a number of other countries, import substitution is becoming one of the strategic objectives of the Russian economy The comparative assessment of standardness and effectiveness of six antimicrobial commercial producer-varying disks for clinical and test strains of microorganisms has been conducted. It is based on analyzing of the variation coefficient (Cv) of inhibition zone diameters, mean values, and diameter values ranges, which are indicators of reproducibility, accuracy and efficiency, respectively. Discs from BD BBL result in insignificant variations of inhibition zone diameters, providing accurate and reproducible data.

Article Synopsis
  • The study investigates the virulence of two clinical strains of the Beijing genotype associated with drug-resistant tuberculosis in C57BL/6 mice, highlighting their correlation to global health issues.
  • Strains 267/47 (pre-XDR) and 120/26 (MDR) showed differing survival rates in infected mice, with 10% and 40% survival, respectively, compared to 70% for the H37Rv strain.
  • Comparison of immune responses revealed significant differences in cytokine gene expression, particularly with downregulated pro-inflammatory and anti-inflammatory genes in mice infected with the more virulent strain 267/47.

The aim of the study is to assess the effectiveness of quality control of transport environments in accordance with the requirements of domestic and international standards using aerobic and optional anaerobic microorganisms. The results of a comparative assessment of the survival of 10 strains of aerobic and optional anaerobic microorganisms after storage in the semi-liquid transport environment of Ames for 24 and 48 hours at temperatures (4-8)0 C and (20-25)0 C in accordance with the requirements of domestic and international standards are presented. The methodology described in ICC 4.

Article Synopsis
  • The Central Asia Outbreak (CAO) clade is a serious public health concern in Central Asia, featuring strains that are both multidrug-resistant and highly transmissible.
  • Research using C57Bl/6 mice revealed that those infected with the drug-resistant Rostov strain died within 48 days, while a more traditional strain, H37Rv, had better survival rates.
  • Analysis of lung and liver tissues showed different pathological effects from the two strains, with the Rostov strain causing a higher bacterial load and severe physical depletion in infected mice.

The results of the comparative tests of the «Agar Muller-Hinton II - Obolensk» nutrient medium developed in SRCAMB, Obolensk, and the control nutrient medium imported «Mueller Hinton II Agar» are presented in the study. The susceptibility of bacterial clinical strains to antimicrobial agents (AMP) was determined by the disc diffusion method and the method of gradient diffusion (E-test). The carbapenemase activity of the strains carrying the carbapenemase genes was determined by CIM-test.

The import substitution becomes one of strategic tasks of Russian economy as a result of imposition of economic sanctions on part of the USA, EU countries, Japan and number of other states. The development of structure and technology of production of national import substituted growth mediums permits satisfying needs of laboratory service of Russia inactive storage and to secure appropriate response to occurring challenges and new biological menaces and support bio-security of state at proper level. The presented data concerning substantiation of nomenclature of growth mediums and transport system permit satisfying in fullness the needs of clinical and sanitary microbiology in growth mediums of national production and to give up of import deliveries without decreasing of quality of microbiological studies.

Real-time polymerase chain reaction was used to develop a one-stage procedure for molecular genetic analysis of Mycobacterium tuberculosis (MBT) DNA in order to determine mutations associated with drug resistance to the antituberculous agents: isoniazid and rifampicin. To analyze the spread of drug-resistance of the causative agent of tuberculosis in Russia, two thousand MBT strains were studied in 24 regions of all the federal districts. Testing 1406 MBT strains isolated by first detected and untreated patients revealed multidrug resistance (MDR) in 21.
