Small-molecule inhibitors of MDM2 that block the MDM2-p53 protein-protein interaction have been considered as potential therapeutic agents for the treatment of cancer. Here, we identify five highly potent inhibitors of MDM2 (termed as WY 1-5) that display significant inhibitory effects on MDM2-p53 interaction by using a combined strategy of pharmacophore modeling, virtual screening, and molecular docking studies. It has been known for many years that the peroxidase activity of cyclooxygenase 1 and 2 (COX-1 and COX-2) can be reactivated in vitro by the presence of phenol, which serves as a reducing compound, but the underlying mechanism is still poorly understood. In the present study, we use phenol as a model compound to investigate the mechanism by which the peroxidase activity of human COXs is reactivated after each catalytic cycle. Molecular docking and quantum mechanics calculations are carried out to probe the interaction of phenol with the peroxidase site of COXs and the reactivation mechanism.

Developing cheap metal nanocatalysts with controllable catalytic activity is one of the critical challenges for improving hydrogen storage in magnesium (Mg). Here, it is shown that the activity of graphene-anchored Co-Ni nanocatalysts can be regulated effectively by tuning their composition and morphology, which results in significantly improved hydrogen storage in Mg. The catalytic activity of supported Co-Ni nanocatalysts is demonstrated to be highly dependent on their morphology and composition.
