To generate a non-red/green fluorescent fusion histone protein in , we have generated a C-terminal mTurquoise2-tagged HIS-72 at the endogenous locus using CRISPR. We found that HIS-72::mTurquoise2 localizes in a similar pattern to the previously published HIS-72::GFP strain.

Although cytokinesis has been intensely studied, the way it is executed during development is not well understood, despite a long-standing appreciation that various aspects of cytokinesis vary across cell and tissue types. To address this, we investigated cytokinesis during the invariant embryonic divisions and found several parameters that are altered at different stages in a reproducible manner. During early divisions, furrow ingression asymmetry and midbody inheritance is consistent, suggesting specific regulation of these events.

Article Synopsis
  • Separase is a critical protease involved in chromosome segregation and membrane trafficking, but its regulation of membrane events is not fully understood.
  • A mutagenesis screen identified 68 lines suppressing a temperature-sensitive separase mutant, with 14 intragenic lines mapping to key residues, showing a relationship between these mutations and separase function.
  • The study also revealed that other mutations in related proteins like PPH-5 and HSP-90 contribute to suppressor activity, paving the way for deeper investigation into separase regulation.
