Staging of pediatric Hodgkin lymphoma is currently based on the Ann Arbor classification, incorporating the Cotswold modifications and the Lugano classification. The Cotswold modifications provide guidelines for the use of CT and MRI. The Lugano classification emphasizes the importance of CT and PET/CT in evaluating both Hodgkin lymphoma and non-Hodgkin lymphoma but focuses on adult patients.

Importance: The current standard-of-care salvage therapy in relapsed/refractory classic Hodgkin lymphoma (cHL) includes consolidation high-dose chemotherapy (HDCT)/autologous stem cell transplant (aSCT).

Objective: To investigate whether presalvage risk factors and fludeoxyglucose-18 (FDG) positron emission tomography (PET) response to reinduction chemotherapy can guide escalation or de-escalation between HDCT/aSCT or transplant-free consolidation with radiotherapy to minimize toxic effects while maintaining high cure rates.

Design, Setting, And Participants: EuroNet-PHL-R1 was a nonrandomized clinical trial that enrolled patients younger than 18 years with first relapsed/refractory cHL across 68 sites in 13 countries in Europe between January 2007 and January 2013.

Background: Diffuse hemispheric glioma, H3G34R/V-mutant (DHG-H3G34) is characterized by poor prognosis and lack of effective treatment options. DHG-H3G34R further harbor deactivation of Alpha-Thalassemia/Mental Retardation Syndrome X-linked protein (ATRX; DHG-H3G34R_ATRX) suggesting a unique interaction of these two oncogenic alterations. In this study, we dissect their cell biological interplay, investigate the impact on telomere stabilization and, consequently, validate a targeted therapy approach.

Germ cell tumors of the testis are the most common tumor entities in young men. Since the introduction of platinum-based chemotherapy in the 1970s, most patients can be cured despite the aggressiveness of germ cell tumors. Optimal serum tumor markers are required for diagnostics, therapy monitoring and aftercare, and these are subject to high requirements.

Testicular germ cell tumours (GCTs) represent a paradigm for the usefulness of serum tumour markers in clinical management of diseases. However, the tumour markers currently in use, beta human chorionic gonadotropin (bHCG), alpha fetoprotein (AFP) and lactate dehydrogenase (LDH) are expressed in less than 50% of GCT cases. In 2011, microRNA-371a-3p (currently named M371) was suggested as a novel marker for the first time.

Article Synopsis
  • Hodgkin lymphoma involving the central nervous system (CNS) is very rare, with a reported incidence of 0.9% in a study of nearly 5,000 pediatric patients.
  • Most cases had single lesions in areas like the thoracic and lumbar spine, which originated from nearby soft tissue or bone rather than directly in the CNS parenchyma.
  • After two cycles of chemotherapy, a significant majority of lesions showed complete metabolic response, and while some lesions were treated with radiation, relapses were minimal, highlighting the importance of surrounding tissues in the management of CNS involvement.

Article Synopsis
  • Brain metastases (BM) are a critical issue for patients with HER2-positive metastatic breast cancer, necessitating better treatment options; the study examined the effectiveness of the antibody-drug conjugate trastuzumab deruxtecan (T-DXd) compared to trastuzumab emtansine.* -
  • In the TUXEDO-1 trial involving patients with HER2-positive BC and active BM, the results showed a median progression-free survival (PFS) of 21 months and an overall survival (OS) that had not been reached at 26.5 months of follow-up, indicating positive outcomes for T-DXd.* -
  • No new safety concerns emerged throughout the trial, with fatigue being the most reported

We report the creation of ultracold ground state ^{6}Li^{40}K polar molecules with high efficiency. Starting from weakly bound molecules, stimulated Raman adiabatic passage is adopted to coherently transfer the molecules to their singlet rovibrational ground state |X^{1}Σ^{+},v=0,J=0⟩. By employing a singlet stimulated Raman adiabatic passage pathway and low-phase-noise narrow-linewidth lasers, we observed a one-way transfer efficiency of 96(4)%.

Article Synopsis
  • Hypodense volumes (HDV) in mediastinal masses were identified in 29.7% of Hodgkin lymphoma patients analyzed from a study involving 1178 staging CT scans.
  • These HDVs were predominantly found in larger tumor volumes and exhibited various patterns, including single lesions in 69.4% of cases and well delineated lesions in 70.1% of cases.
  • Patients with HDV displayed more severe symptoms and had a lower 5-year progression-free survival rate (79.6% for HDV > 40 ml) compared to those with lower HDV levels, indicating a potential connection between HDV presence and poorer prognosis in Hodgkin lymphoma.

Background And Objective: Serum levels of microRNA-371a-3p (M371) represent a novel and sensitive biomarker of germ cell tumours (GCTs). This study analysed the utility of M371 to identify viable cancer (VC) in postchemotherapy (pc) residual masses with the underlying goal of avoiding overtreatment.

Methods: A multicentric, prospective diagnostic study was conducted in 180 GCT patients undergoing pc resection of residual masses.

Introduction: Paraneoplastic hyperthyroidism (PH) has been reported in patients with testicular germ cell tumors (GCTs), sporadically. This disorder is caused by extremely elevated serum levels of beta-human chorionic gonadotropin (bHCG). To date, little is known about the prevalence of PH, and its clinical features are poorly understood.

Background: Sex cord gonadal stromal tumors compose less than 10% of all testicular neoplasms and consist of a variety of histological subtypes. In 2016, the World Health Organization introduced a novel subtype, the myoid gonadal stromal tumor, that consists of spindle-shaped cells with immunohistologic features of muscle cells. Only few cases have been reported to date.

Article Synopsis
  • New lung lesions were identified in 9.2% of pediatric Hodgkin lymphoma patients after two chemotherapy cycles, indicating that these may not be related to cancer progression.
  • The study utilized interim CT scans to differentiate between true lung metastases and benign lung lesions, using guidelines from the Fleischner glossary for classification.
  • Most newly detected pulmonary nodules were small (under 10 mm) and often regressed by the time of later evaluations, suggesting they were likely not harmful manifestations associated with the original lymphoma.

Article Synopsis
  • * Out of 258 patients monitored over 18 months, the M371 test achieved 100% sensitivity and 96.3% specificity for relapse detection, with some ability to detect relapses earlier.
  • * While the M371 test shows promise for routine use in monitoring relapses, postoperative levels of M371 did not predict future relapses, and the time advantage for earlier detection was not significant.

We present the comparison of a field-programmable-gate-array (FPGA) based digital servo module with an analog counterpart for the purpose of laser frequency stabilization to a high-finesse optical cavity. The transfer functions of both the digital and analog modules for proportional-integral-derivative control are measured. For the lasers stabilized to the cavity, we measure the singe-sideband power spectral density of fast phase noise by means of an optical beat with filtered light transmitted through the cavity.
