The use of Vero cells for rabies vaccine production was recommended from the WHO in 2005. A controlled production process is necessary to reduce the risk of contaminants in the product. One step towards this is to turn away from animal-derived components (e.

A proteomic approach was used to investigate the dynamic cellular host cell response induced by influenza virus infection in two different vaccine production cell lines, MDCK and Vero. For identification of proteins possibly involved in global host cell response mechanisms and virus-host cell interactions in these producer cell lines, quantitative 2-D DIGE and nanoHPLC-nanoESI-MS/MS analysis were performed. In particular, host cell proteome alterations caused by infection with influenza virus variants showing differences in replication characteristics in MDCK cells were compared.

Article Synopsis
  • A quantitative RT-qPCR assay was developed to analyze how the influenza A virus transcribes and replicates in mammalian cell culture by distinguishing between different types of viral RNA.
  • Synthetic viral RNAs were created for accurate validation and measurement of the assay, which showed high sensitivity and consistency in results.
  • The study also observed that during early infection phases in MDCK cells, mostly viral messenger RNAs were produced first, followed by replicative intermediates and viral genomes, indicating a specific timing in the replication process.

Over the last years virus-host cell interactions were investigated in numerous studies. Viral strategies for evasion of innate immune response, inhibition of cellular protein synthesis and permission of viral RNA and protein production were disclosed. With quantitative proteome technology, comprehensive studies concerning the impact of viruses on the cellular machinery of their host cells at protein level are possible.
