Oligonucleotide Microchip for the Identification of Infectious Agents of Reproductive System with Simultaneous Analysis of Determinants of Resistance to Antimicrobial Substances.

We developed a multiplexed DNA microarray-based assay allowing identification of 12 causative agents of reproductive tract infections with the simultaneous detection of 47 genetic determinants of resistance to antimicrobial substances. The microarray was tested on 93 isolates of Neisseria gonorrhoeae, 32 isolates of Treponema pallidum and 29 samples of Ureaplasma spp./Mycoplasma spp.

EXPERIENCE OF APPLICATION OF VOLUME FORMING AGENT «DAM+» IN TREATMENT OF ANAL INCONTINENCE ASSOCIATED WITH INCOMPETENCE AND TRAUMA OF ANAL SPHINCTER.

The attempts of introduction of volume forming agent in submucous layer started at the beginning of 1990th. The aim of these innovations was to rise of basal pressure. This research has been performing since 2007.

Characteristics of cardiac cell cultures derived from human myocardial explants.

Primary cell cultures derived from human myocardial explants were obtained and characterized. The explant cultures contained cardiac stem cells (c-kit(+); ≈ 4%), microvascular cells (endothelial cells and pericytes), fibroblasts, and myofibroblasts. It was demonstrated that culturing of cardiac cells in cardiospheres did not promote enrichment of the cell culture with stem cells.

[The changes in the brain's electrical activity in children with cerebral palsy during the complex rehabilitation].

One hundred and five children, aged from 3 to 7 years, with the diagnosis "spastic diplegia cerebral palsy" were treated. Patients were stratified into three groups: group I (n=36) received three courses of microcurrent therapy (MENS) in addition to standard treatment; group II (n=38) received three courses of MENS in the combination with two treatment courses with the nootropic drug cortexin; children of group III (n=31) received standard therapy using massage and gymnastics. MENS was carried out in courses, including 15 sessions each, using the apparatus "MEKS".

[The role of microcurrent reflexotherapy in combination with neuroprotector in the rehabilitation of the patients with infantile cerebral palsy].

The objective of the present study was to estimate the therapeutic efficacy of the combined treatment of infantile cerebral palsy by means of microcurrent reflexotherapy (MCRT) in combination with the neuroprotector cortexin. This treatment including 15 sessions with the use of a MAKS apparatus was given to 69 children at the age from 3 to 7 years. They were randomly allocated to two groups.

[Method for automated extraction and purification of nucleic acids and its implementation in microfluidic system].

A method and a microfluidic device for automated extraction and purification of nucleic acids from biological samples have been developed. The method involves disruption of bacterial cells and/or viral particles by combining enzymatic and chemical lysis procedures followed by solid-phase sorbent extraction and purification of nucleic acids. The procedure is carried out in an automated mode in a microfluidic module isolated from the outside environment, which minimizes contact of the researcher with potentially infectious samples and, consequently, decreases the risk of laboratory-acquired infections.

[Antianemic effect of reamberin in rats with acute alloxan-induced diabetes].

The influence of reamberin on hematological indices in the acute period of alloxan-induced diabetes has been studied. It is established that anemia develops within 72 hours after alloxan injection at a diabetogenic dose in rats, which is related to bone marrow erythropoiesis suppression and is accompanied by inflammatory leukocytosis. Single administration of reamberin in a dose equivalent to the human therapeutic dose produces correction of the leukocyte content in the blood, normalizes the bone marrow erythropoiesis, and prevents anemia development in rats with alloxan-induced diabetes.

[Development of a biochip for quantitative determination of two forms of prostate specific antigen using internal calibration curve].

Three-dimensional gel-based microchip allowing simultaneous quantitative detection of total (PSAtot) and free (PSAfree) forms of prostate specific antigen in human serum (in a format "one patient-one biochip") was developed. A method, which doesn't require preliminary construction of calibration curves when performing an assay, was applied for quantitative determination of PSAtot and PSAfree. Gel elements with immobilized antigen (PSA) in different concentration, forming an internal calibration curve, were included in a structure of the microchip, in addition to the elements with immobilized antibodies specific against PSAtot and PSAfree.

[Chromatin modifications during X-chromosome inactivation in female mammals].

In female mammals, the process of dosage compensation occurs during early embryonic development. As a result of this, one of X-chromosomes becomes transcriptionally inactive. This process is accompanied by chromatin remodeling on inactivated X-chromosome, providing transcriptional silencing of the genes and maintenance of their inactive state.

[Antigenic specificity of recombinant polypeptides, containing fragments of hepatitis E virus proteins].

Antigenic specificity of recombinant polypeptides HE40 and HE60 containing fragments of gene ORF2 and ORF3 protein products of hepatitis E, strain Burma, produced in E. coli cells, is analyzed. Blood sera from patients with acute hepatitis from an endemic region in Uzbekistan were tested for IgG to recombinant antigens by solid-phase enzyme immunoassay with a protein fragment coded by PRF3 gene, a synthetic peptide previously characterized in a commercial test system, as the positive control.

[Antigenic activity of delta-antigen peptides corresponding to three genotypes of the delta hepatitis virus].

Six synthetic peptides from immunodominant region 65-80 aa derived from the sequences of delta-antigen corresponding to 3 HDV genotypes were obtained. Their antigenic activities were assessed by indirect enzyme immunoassay with sera of patients with hepatitis D and of asymptomatic carriers of anti-delta antibodies. Differences in antigenic characteristics of full-size 65-80 and truncated 71-80 aa peptides corresponding to three HDV genotypes were revealed.

[Analysis of the organization of immunodominant epitopes of hepatitis virus E orf2 and orf3 proteins using synthetic peptides].

A set of synthetic peptides of different length derived from proteins encoded by open reading frames (ORF) 1, 2, and 3 of hepatitis E virus (HEV) genome was synthesized and tested for antigenicity in indirect enzyme immunoassay. Peptides 414-433 (ORF2 protein) and 99-119 (ORF3) specifically react with antiHEV-positive sera collected during outbreaks in the Central Asian areas of the former USSR. The antigenic structure of the Central Asian variants and the Burmese types of HEV was found similar.

[The ATP content of the neutrophils and whole blood in the inhabitants of regions in the Altai Territory subjected to nuclear testing exposure].

The bioluminescent method was used in the studies of the influence of ionizing irradiation and/or xenobiotics on the content of ATP in RBC and neutrophils of rats, and in whole blood and neutrophils of 80 examined women of Altai Region exposed to ionizing radiation during a series of nuclear tests in Semipalatinsk in 1949-1965. Deviations from the normal ATP content were measured with due to account of the natural variability of a given metabolite. For rats, deviations from the content of ATP in erythrocytes were short-term, those in neutrophils were long-term.

Physico-chemical properties of the recombinant L. mingrelica luciferase synthesized by E. coli cells have been studied.

[A comparative study of the expression of activation markers on T-lymphocyte subpopulations in different forms of bronchial asthma].

In this study we investigated a phenotype of intact and PHA-stimulated T-lymphocytes from peripheral blood of patients with allergic and nonallergic asthma. Expression of the activation markers (CD25, CD71, HLA-DR) on subsets of T-lymphocytes (CD4+ and CD8+) was determined using double-colour flow cytometry. It was found that nonallergic patients had increased percentages of CD4+CD25+, CD8+CD25+, CD4+HLA-DR+, CD8+HLA-DR+ cells.

[Synthesis of platelet activation factor analogs].

Novel structural analogues of the platelet activating factor (PAF), rac-1-octadecyl-2-allyl-glycerol-3-phosphocholine and rac-1-octadecyl-2-allyl-1-thioglycerol-3-phosphocholine , have been synthesized. The phospholipids obtained showed neither PAF-agonistic nor PAF-antagonistic activity at the concentration of 10(-5) to 10(-7) M.

[Effect of flanking regions on the expression of the human interleukin-2 chromosomal gene in a murine myeloma cell line].

A human interleukin-2 (IL-2) gene was isolated from genomic DNA library. The isolated gene with 5'- and 3'-flanking sequences of various lengths was inserted into plasmids derived from the retroviral vector pPSneo. The recombinant plasmids were transfected into myeloma X63Ag8-653 cells.

[Enzyme diagnosis of mechanical jaundice].

Combined enzymodiagnostic program--estimation of total activity of blood serum enzymes aspartate aminotransferase (AST), alanine aminotransferase (AAT), alkaline phosphatase (AP), gamma-glutamyl transferase (gamma-GT), glutamate dehydrogenase (GDH), acetyl cholinesterase (ACE), butyryl cholinesterase (BCE)--was studied to evaluate its diagnostic validity in surgical clinic. In mechanical jaundice of various origin mean values of the enzymatic activities studied as well as the enzymatic coefficients were distinctly altered as compared with control values: AST/AAT, gamma-GT/AST as well as the newer coefficients BCE/ACE, AP/gamma-GT, (formula; see text). The jaundices of tumoral and non-tumoral genesis caused markedly dissimilar alterations in the coefficients AST/AAT and (formula; see text).

[Enzymological evaluation of the hepatotoxicity of ampicillin and its therapeutic form, roscillin, in the treatment of pyelonephritis in pregnancy].

The combined enzymological investigation including determination of the total activity of asparagine transaminase and alanine transaminase, two serum enzymes, alkaline phosphatase, gamma-glutamyl transpeptidase, acetyl cholinesterase, and butyryl cholinesterase was applied to two groups of pregnant women with pyelonephritis treated with ampicillin (12 patients) and roscillin (14 patients). The investigation was performed at the following stages: before the treatment, on the 7th and on the 12th day of the treatment. No statistically significant differences in the average values of the activity of the above enzymes at these stages were observed in patients of the both groups which indicated the absence of the hepatotoxic effect of the preparations on the patients of a group as a whole.
