Lineage switching can induce therapy resistance in cancer. Yet, how lineage fidelity is maintained and how it can be lost remain poorly understood. Here, we have used CRISPR-Cas9-based genetic screening to demonstrate that loss of SMARCB1, a member of the SWI/SNF chromatin remodeling complex, can confer an advantage to clear cell renal cell carcinoma (ccRCC) cells upon inhibition of the renal lineage factor PAX8.

Article Synopsis
  • - Neuroendocrine prostate cancer (NEPC) often develops from typical prostate cancer following treatments like androgen deprivation therapy, leading to a process called neuroendocrine transdifferentiation that results in tumors with poor survival rates.
  • - Researchers analyzed 54 prostate cancer samples over 10 years to differentiate between NE-like tumors and standard adenocarcinoma, identifying specific gene and miRNA expressions that characterize NEPC.
  • - A predictive gene expression signature for NEPC was created, highlighting molecular markers that could help enhance diagnosis and treatment options for this challenging cancer subtype.
