Most eukaryotic genes encode polypeptides that are either obligate members of hetero-stoichiometric complexes or clients of organelle-targeting pathways. Proteins in these classes can be released from the ribosome as "orphans"-newly synthesized proteins not associated with their stoichiometric binding partner(s) and/or not targeted to their destination organelle. Here we integrate recent findings suggesting that although cells selectively degrade orphan proteins under homeostatic conditions, they can preserve them in chaperone-regulated biomolecular condensates during stress.

Background: A novel image-based method (AgileBrain) demonstrates construct validity as a measure of wellbeing in the general working adult population.

Method: Analysis of data from four large nationally representative samples of American full-time workers employed by mid-to-large size companies conducted in November 2021 ( = 812), May 2022 ( = 810), June 2023 ( = 986), and January 2024 ( = 1,179).

Results: Across all four studies, AgileBrain demonstrates convergent validity across multiple established indicators of subjective wellbeing including the Patient Health Questionnaire (PHQ-9), Center for Epidemiological Studies Depression Scale (CESD-10), Generalized Anxiety Disorder Scale (GAD-7), Neuroticism (BFI-S), UCLA Loneliness Scale, Perceived Stress Scale (PSS-10), Coping Styles (Brief COPE-28), self-reported diagnosed neurodiversity conditions and symptoms, and trauma history.

Previous research has demonstrated that interpersonal dynamics are fractal, and that conflict is a key control parameter that drives fractal complexity. The present study aimed to extend this line of research to examine the putative fractal structure of conflict dynamics over time, and the role that this self-organizing fractal structure may play in the resilience of romantic relationships. An experience sampling methodology was used to assess levels of conflict, satisfaction, and commitment in the dating relationships of undergraduate students, three times per day for 30 days.

This introduction to a special issue of Nonlinear Dynamics, Psychology and Life Sciences on the topic of resilience discusses the contributing articles in terms of their flexibility in methods, models, scale, and contexts combined with their integrity in shared theoretical understanding and generative knowledge. The ubiquity of resilience is discussed, a feature of potentially any living or non-living system and substance. This breadth calls for a flexible set of models and methods, along with the quest for integrative theory to make resilience science more resilient.

The heat shock response (HSR) is a gene regulatory program controlling expression of molecular chaperones implicated in aging, cancer, and neurodegenerative disease. Long presumed to be activated by toxic protein aggregates, recent work suggests a new functional paradigm for the HSR in yeast. Rather than toxic aggregates, adaptive biomolecular condensates comprised of orphan ribosomal proteins (oRP) and stress granule components have been shown to be physiological chaperone clients.

Ribosome biogenesis is among the most resource-intensive cellular processes, with ribosomal proteins accounting for up to half of all newly synthesized proteins in eukaryotic cells. During stress, cells shut down ribosome biogenesis in part by halting rRNA synthesis, potentially leading to massive accumulation of aggregation-prone 'orphan' ribosomal proteins (oRPs). Here we show that, during heat shock in yeast and human cells, oRPs accumulate as reversible peri-nucleolar condensates recognized by the Hsp70 co-chaperone Sis1/DnaJB6.

Background: A unified model of human motivation has been recently introduced that integrates all prior "mini-theories" of motivation into a single, symmetrical model based on first principles: four life domains crossed by three levels of attainment, resulting in 12 discrete motivations. Evidence from a series of studies using a novel image-based method is used to test structural hypotheses derived from a unified model of human motivation.

Method: The studies employ large samples (810n to 986n) of working adults who conducted a time-constrained image-based exercise to measure the relative presence or absence of different emotional needs.

Temperature increases as light intensity rises, but whether light signals can be directly linked to high temperature response in plants is unclear. Here, we find that light pre-treatment enables plants to survive better under high temperature, designated as light-induced thermotolerance (LIT). With short-term light treatment, plants induce light-signaling pathway genes and heat shock genes.

Revisions are proposed to the taxonomic model of human motivation of Forbes (Review of General Psychology, 15(2), 85-98, 2011) in order to incorporate a heretofore missing fourth life domain, the spiritual. The growing literature on spiritual motives is systematically reviewed in accordance with literature review standards for theory development (Templier & Paré, 2018) focusing on the objective of identifying comprehensive theoretical systems that explicitly incorporate the spiritual domain as one of a limited set of human life domains. The structure of the Forbes model is contrasted with thirteen theoretical systems that explicitly incorporate the spiritual as a fourth life domain.

Ten good outcome and ten poor outcome psychotherapy cases were compared to investigate whether or not the temporal stability and flexibility of their process variables can predict their outcomes. Each participant was monitored daily using the Therapy Process Questionnaire (TPQ), which has 43 items and seven sub-scales, and responses over time were analyzed in terms of correlation robustness and correlation variability across the TPQ sub-scales. "Correlation robustness" and "correlation variability" are two basic characteristics of any correlation matrix: the first is calculated as the sum of the absolute values of Pearson correlation coefficients, the second as the standard deviation of Pearson correlation coefficients.
