Background: Chronic active Epstein-Barr virus (CAEBV) infection is a severe, life-threatening condition characterized by persistent Epstein-Barr virus (EBV) infection and the clonal expansion of infected T or NK cells, leading to systemic inflammation, organ damage, and complications such as hemophagocytic lymphohistiocytosis and lymphoma. Allogeneic hematopoietic stem cell transplantation (HSCT) is the only effective treatment for eradicating EBV-infected cells; however, donor availability is limited. Fulminant myocarditis (FM) is a severe inflammatory condition of the myocardium that often results in sudden death, particularly in young individuals. In this study, we employed single-nucleus and spatial transcriptomics to perform a comprehensive analysis of coxsackievirus B3 (CVB3)-induced FM in A/J mice, spanning seven distinct time points pre- and post-treatment. Our findings reveal that mesothelial cells play a critical role in the early stage of myocarditis by acting as primary targets for CVB3 infection.

We describe a previously uncharacterized ATP-binding cassette A1 super enhancer RNA (ABCA1-seRNA)-mediated cholesterol efflux. In addition, it promoted macrophage inflammatory cytokine release, and was causally correlated with coronary artery disease severity. Mechanistically, ABCA1-seRNA upregulated cholesterol efflux by interacting with mediator complex subunit 23 and recruiting retinoid X receptor-alpha and liver X receptor-alpha to promote ABCA1 transcription in a manner.
