Acetaminophen is a common over-the-counter medication that recently gained substantial media attention regarding its use by pregnant individuals. In this clinical perspective, we discuss the strengths and limitations of the published literature on the effect of maternal acetaminophen use in pregnancy on the child's risk of developing attention-deficit and hyperactivity disorder (ADHD) and autism spectrum disorder (ASD). Studies included were specifically selected on the basis of the quality and validity of ADHD or ASD outcome definitions.

Article Synopsis
  • The study aimed to explore whether certain genetic variants (SNPs) related to taxane chemotherapy affect breast cancer patients' need for social benefits after treatment.
  • Researchers analyzed data from premenopausal women diagnosed with breast cancer, focusing on their receipt of health and labor market-related benefits over a five-year period.
  • The findings indicated that there was no significant link between the selected SNPs and the likelihood of receiving social benefits post-chemotherapy.

  • The European Medicines Agency (EMA) is recommending precautionary measures due to concerns about neurodevelopmental disorders in children of men treated with valproate.
  • The UK's Medicines and Healthcare products Regulatory Agency (MHRA) went further, advising against prescribing valproate to anyone under 55.
  • Members of ENTIS and OTIS argue that these warnings are premature and that the scientific evidence does not strongly support the risk of paternal transmission of issues from valproate.

Aim: We aimed to assess if dicloxacillin/flucloxacillin reduces the therapeutic efficacy of direct oral anticoagulants (DOACs) and the underlying molecular mechanism.

Methods: In a randomized, crossover study, we assessed whether dicloxacillin reduces oral absorption of drugs through P-glycoprotein (P-gp) during 10 and 28 days of treatment. To study the impact of dicloxacillin/flucloxacillin on intestinal and hepatic expression of P-gp in vitro, we usd LS174T cells and 3D spheroids of primary human hepatocytes.

Objective: Patients with Osteogenesis Imperfecta (OI) have varying degrees of bone fragility and increased fracture rates. There is a paucity of data related to complications to pregnancies in patients with OI and to their offspring. With this study we aim to evaluate the risk of complications to pregnancies, delivery, and offspring in pregnancies where the mother or father have OI.

Background: The prevalence of diabetes and coexisting multimorbidity rises worldwide. Treatment of this patient group can be complex. Providing an evidence-based, coherent, and patient-centred treatment of patients with multimorbidity poses a challenge in healthcare systems, which are typically designed to deliver disease-specific care.

Article Synopsis
  • - A study was conducted to assess substance use (including alcohol and nicotine) in pregnant women in Denmark, as there is limited verified data on this topic.
  • - The research involved urine samples and questionnaires from nearly 1900 pregnant women attending ultrasound scans; results showed that 25% tested positive for substances, with a confirmed prevalence of 10.8% for all substances, predominantly nicotine usage.
  • - The findings indicated that while self-reporting of nicotine was high, reporting for alcohol and cannabis was notably low, highlighting discrepancies between self-reported data and actual substance use detected through screening.

The aim of this study was to examine variations in use of antidepressants among children and adolescents in the three Scandinavian countries (Sweden, Norway, and Denmark). We identified new users of antidepressants (5-17 years) during 2007-2018 and described the annual incidence rate, treatment duration, concomitant psychotropic drug use, and the clinical setting of the prescribing physician (in Sweden and Denmark). Incident use of antidepressants increased by a factor 1.

Background And Aim: Paternal use of analgesics during the time of conception and adverse birth outcomes are poorly studied. We investigated the association between paternal exposure to non-steroid anti-inflammatory drugs and opioids within 3 months before the date of conception and the risk of adverse birth outcomes (preterm birth, small for gestational age, low Apgar score, and major congenital malformations).

Methods: We used nationwide data from the Danish health registers.

Article Synopsis
  • The study aimed to analyze opioid prescription patterns among pregnant women in Denmark from 1997 to 2016, utilizing national health care registers.
  • Results indicated stable opioid usage rates among women with live births, but revealed a decline in codeine use and a significant increase in tramadol prescriptions.
  • Factors such as lower socioeconomic status, older age, obesity, and smoking were associated with higher odds of opioid use during pregnancy.

Exposure to certain medications can disrupt processes of fetal development, including brain development, leading to a continuum of neurodevelopmental difficulties. Recognizing the deficiency of neurodevelopmental investigations within pregnancy pharmacovigilance, an international Neurodevelopmental Expert Working Group was convened to achieve consensus regarding the core neurodevelopmental outcomes, optimization of methodological approaches and barriers to conducting pregnancy pharmacovigilance studies with neurodevelopmental outcomes. A modified Delphi study was undertaken based on stakeholder and expert input.

Background: Serotonin syndrome is a potentially life-threatening syndrome with manifestations spanning from mild adverse effects to life-threatening toxicity. The syndrome is caused by overstimulation of serotonin receptors by serotonergic drugs. Since the use of serotonergic drugs is increasing, primarily due to the widespread use of selective serotonin reuptake inhibitors, cases of serotonin syndrome have likely seen a parallel increase.

Objective: To compare the occurrence of sudden sensorineural hearing loss following immunization with BNT162b2 (Comirnaty®; Pfizer BioNTech) or mRNA-1273 (Spikevax®; Moderna) to the occurrence among unvaccinated individuals.

Study Design: Cohort study.

Setting: Nationwide Danish health care registers comprised all Danish residents living in Denmark on October 1, 2020, who were 18 years or older or turned 18 in 2021.

Flucloxacillin is a widely used antibiotic. It is an agonist to the nuclear receptor PXR that regulates the expression of cytochrome P450 (CYP) enzymes. Treatment with flucloxacillin reduces warfarin efficacy and plasma concentrations of tacrolimus, voriconazole, and repaglinide.

Background: In 2020, the European Medicines Agency recommended testing patients for dihydropyrimidine dehydrogenase (DPD) deficiency before systemic treatment with fluoropyrimidines (FP). DPD activity testing identifies patients at elevated risk of severe FP-related toxicity (FP-TOX). The two most used methods for DPD testing are DPYD genotyping and DPD phenotyping (plasma uracil concentration).

Purpose: Breast cancer treatment is associated with adverse effects, which may delay return-to-work. Single nucleotide polymorphisms (SNPs) may influence the risk and severity of treatment toxicities, which in turn could delay return-to-work. We examined the association of 26 SNPs with return-to-work in premenopausal women with breast cancer.

Gabapentin and pregabalin have been associated with an increased risk of fragility fractures. Due to differences in pharmacokinetics, we aimed to assess the fracture-risk difference between the two medicines. We performed a Danish nationwide new user, high-dimensional propensity score-matched cohort study to assess the 90-day risk of fragility fractures among adults, from January 1996 to December 2018.
