DnaJ heat shock protein family member C5 beta (DNAJC5B), also known as cysteine-string protein beta, exhibits a prominent expression in testicular tissue and plays an important role in acrosomal exocytosis in vitro. Nevertheless, the precise role and underlying mechanism of DNAJC5B in spermatogenesis and male fertility remain poorly understood. The meta-analysis of RNA-sequencing datasets from porcine and murine testes reveals that Dnajc5b could be a pivotal factor in spermatogenesis.

Polynyas play a critical role in the formation of Antarctic Bottom Water and the enhancement of polar primary productivity. Accurate and exhaustive identification of Antarctic polynyas is fundamental to advancing in-depth research. However, due to methodological limitations, previous studies paid more attention to frequent polynyas and infrequent polynyas have not been investigated much despite that they could be vulnerable to climate change.

Objective: Training data fuel and shape the development of artificial intelligence (AI) models. Intensive data requirements are a major bottleneck limiting the success of AI tools in sectors with inherently scarce data. In health care, training data are difficult to curate, triggering growing concerns that the current lack of access to health care by under-privileged social groups will translate into future bias in health care AIs.

Article Synopsis
  • The summer Eurasian westerly jet is becoming weaker and wavier, leading to more frequent weather extremes, though the main cause is still under debate.
  • A significant 140% increase in interannual variability of the East Asian jet (EAJ) since the late 20th century is linked to changes in large-scale circulation patterns across Eurasia, resulting in notable climate impacts like heatwaves and monsoon shifts.
  • The Scandinavian pattern from February plays a major role in this variability, suggesting a connection between oceanic and atmospheric conditions, although advanced climate models don't show similar circulation changes, indicating that much of the EAJ's recent variability may be due to natural fluctuations rather than human activity.

The downslope plumes of dense shelf water (DSW) are critical for the formation of Antarctic Bottom Water (AABW), and thus to the exchange of heat and carbon between surface and abyssal ocean. Previous studies have shown that tides and overflow-forced topographic Rossby waves (TRWs) may have strong impact on the downslope transport of DSW, but it remains unclear how the combined action of these two processes influence the descent processes of DSW, and of the resulting AABW properties. Here, with a synthesis of historical in situ observations and a set of numerical model experiments, we show that tides and TRWs play comparable roles in AABW formation: they both act to accelerate DSW descent to the abyss, leading to the formation of colder and denser AABW.

The edge of a monsoon region is usually highly sensitive to climate change. Pakistan, which is located on the northern edge of the Indian monsoon, is highly vulnerable to heavy rainfall and has witnessed several debilitating floods exacerbated by global warming in recent years. However, the mechanisms for the frequent Pakistan floods are yet not fully understood.

Ensemble Kalman filter-based targeted observation is one of the best methods for determining the optimal observational array for oceanic buoy deployment. This study proposes a new algorithm suitable for a 'cross-region and cross-variable' approach by introducing a projection operator into the optimization process. A targeted observational analysis was conducted for El Niño-Southern Oscillation (ENSO) events in the tropical western Pacific for the Tropical Pacific Observation System (TPOS) 2020.

Covalent inhibitors of the papain-like protease (PLpro) from SARS-CoV-2 have great potential as antivirals, but their non-specific reactivity with thiols has limited their development. In this report, we performed an 8000 molecule electrophile screen against PLpro and identified an α-chloro amide fragment, termed compound 1, which inhibited SARS-CoV-2 replication in cells, and also had low non-specific reactivity with thiols. Compound 1 covalently reacts with the active site cysteine of PLpro, and had an IC50 of 18 μM for PLpro inhibition.

This article reports the synthesis and characterization of a novel self-immolative linker, based on thiocarbonates, which releases a free thiol upon activation enzymes. We demonstrate that thiocarbonate self-immolative linkers can be used to detect the enzymes penicillin G amidase (PGA) and nitroreductase (NTR) with high sensitivity using absorption spectroscopy. Paired with modern thiol amplification technology, the detection of PGA and NTR were achieved at concentrations of 160 nM and 52 nM respectively.

Article Synopsis
  • Prostate cancer (PCa) poses a serious risk to older men, often progressing to castration-resistant PCa (CRPC) within two years, yet its molecular mechanisms remain largely unknown.
  • The study focused on CKLF-like Marvel transmembrane domain containing family member 5 (CMTM5) and explored its role in PCa by analyzing tissue samples and conducting various in vitro assays with CMTM5-overexpressing cells.
  • Results indicated that CMTM5 is significantly downregulated in PCa tissues, while EGFR is upregulated; overexpression of CMTM5 inhibits cell proliferation, migration, and invasion, while promoting apoptosis, suggesting a potential regulatory pathway involving PI3K/AKT.
