Methods: Patients > 18 years of age, English or Spanish speakers, referred to PT were invited to complete an online survey. Factors that may influence PT satisfaction were examined for patients who attended PT.
Background: Cerliponase alfa is a recombinant human tripeptidyl peptidase 1 (TPP1) enzyme replacement therapy for the treatment of neuronal ceroid lipofuscinosis type 2 (CLN2 disease), which is caused by mutations in the TPP1 gene. We aimed to determine the long-term safety and efficacy of intracerebroventricular cerliponase alfa in children with CLN2 disease.
Methods: This analysis includes cumulative data from a primary 48-week, single-arm, open-label, multicentre, dose-escalation study (NCT01907087) and the 240-week open-label extension with 6-month safety follow-up, conducted at five hospitals in Germany, Italy, the UK, and the USA.

Reaction of Bn-tacn (1-benzyl-1,4,7-triazacyclononane) with paraformaldehyde and PhP(OR) (R = Me or Et) in refluxing THF, followed by acid hydrolysis, yields the macrocyclic (phosphinic acid) derivative, H(Bn-NODP) (1-benzyl-4,7-phenylphosphinic acid-1,4,7-triazacyclononane), which is isolated as its protonated form, H(Bn-NODP)·2HCl·4HO, at low pH (HCl), its disodium salt, Na(Bn-NODP)·5HO at pH 12 (NaOH), or the neutral H(Bn-NODP) under mildly basic conditions (EtN). A crystal structure of H(Bn-NODP)·2HCl·HO confirmed the ligand's identity.
