The aim of this study was to determine the microclimate of brown bear dens depending on their size, their status (active or inactive), and the location of the den. The study included five dens of different sizes and locations in the Velebit Mountains, in the Dinaric Alps. View Article and Find Full Text PDFThe procedure illustrated in this paper represents a new method for transcriptome analysis by PCR (Polymerase Chain Reaction), which circumvents the need for elimination of potential DNA contamination. Compared to the existing methodologies, our method is more precise, simpler and more reproducible because it preserves the RNA's integrity, does not require materials and/or reagents that are used for elimination of DNA and it also reduces the number of samples that should be set up as negative controls. This novel procedure involves the use of a specifically modified primer during reverse transcription step, which contains mismatched bases, thus producing cDNA molecules that differ from genomic DNA.

View Article and Find Full Text PDFDespite being adapted to a wide range of environmental conditions, the vitality of European beech is expected to be significantly affected by the projected effects of climate change, which we attempted to assess with foliar nutrition and crown defoliation, as two different, yet interlinked vitality indicators. Based on 28 beech plots of the ICP Forests Level I network, we set out to investigate the nutritional status of beech in Croatia, the relation of its defoliation and nutrient status, and the effects of environmental factors on this relation. The results indicate a generally satisfactory nutrition of common beech in Croatia.
