Pathogenic gene dysregulation can be attributed to chromatin state change that pre-transcriptionally regulates expression. Recent breakthroughs elucidating the rules governing this DNA control layer, an epigenetic code, unlock a modality in precision medicine to target gene dysregulation across myriad diseases. Here we present a modular platform to design programmable mRNA therapeutics, Epigenomic Controllers (EC), that control gene expression through directed epigenetic change.

This study investigated the added value of using maximum-intensity projection (MIP) images for fully automatic segmentation of lesions using deep learning (DL) in [F]FDG and [Ga]Ga-prostate-specific membrane antigen (PSMA) PET/CT scans. We used 489 staging [F]FDG PET/CT scans from patients diagnosed with melanoma, lymphoma, or lung cancer (391 scans for training and 98 for internal testing). As an external test set, 117 staging [F]FDG PET/CT scans from lymphoma patients (another center, 2 scanners) were used.

Aims: Fam-trastuzumab deruxtecan-nxki (T-DXd) was recently approved for advanced stage or metastatic solid tumours with human epidermal growth factor receptor 2 (HER2) immunohistochemical (IHC) 3+ staining. Data on HER2 IHC testing and knowledge of genomic correlates in lung cancer are scarce. This study analyses genomic characteristics of HER2-expressing tumours and addresses issues with preanalytical variables for lung cancer specimens.

Haskap () berry is rich in anthocyanins, particularly cyanidin-3--glucoside (C3G). In this investigation, a response surface methodology was applied to optimize the anhydrous ethanol-based extraction parameters to obtain the maximum yield of anthocyanins from haskap berry and to compare the recovery of anthocyanins from different extraction methods. The central composite design was employed to study the effect of three independent variables (X = ultrasonic bath power, X = extraction temperature, and X = extraction time) which were found to significantly affect the response variable total anthocyanin content (TAC) and fit to the second-order polynomial model.
