Beta-arrestins (βarrs) are key regulators and transducers of G-protein coupled receptor signaling; however, little is known of how βarrs communicate with their downstream effectors. Here, we use cryo-electron microscopy to elucidate how βarr1 recruits and activates non-receptor tyrosine kinase Src. βarr1 binds Src SH3 domain via two distinct sites: a polyproline site in the N-domain and a non-proline site in the central crest region.

Small GTPases comprise a superfamily of over 167 proteins in the human genome and are critical regulators of a variety of pathways including cell migration and proliferation. Despite the importance of these proteins in cell signaling, a standardized approach for controlling small GTPase activation within living cells is lacking. Herein, we report a split-protein-based approach to directly activate small GTPase signaling in living cells.

There is now wide spread interest in the application of small field of view gamma cameras for clinical use in operating theatres, intensive care units and bedside imaging. The development of these imaging systems some of which have improved spatial resolution has necessitated a reappraisal of the suitability of conventional phantoms intended for use with gamma cameras. The testing of imagers having increased system resolution requires phantoms and test objects of smaller dimensions.
