Microrobots are poised to transform biomedicine by enabling precise, noninvasive procedures. However, current magnetic microrobots, composed of solid monolithic particles, present fundamental challenges in engineering intersubunit interactions, limiting their collective effectiveness in navigating irregular biological terrains and confined spaces. To address this, we design hierarchically assembled microrobots with multiaxis mobility and collective adaptability by engineering the potential magnetic interaction energy between subunits to create stable, self-reconfigurable structures capable of carrying and protecting cargo internally.

We developed a design strategy focusing on pivotal secondary structural motifs-α-helix, β-strand, and β-turn-critical for PPI recognition, using a common core skeleton. The resulting peptide-inspired pyrimidodiazepine scaffolds were further subjected to comprehensive phenotypic screening to evaluate their efficacy. Our strategy offers a transformative approach to developing small-molecule PPI modulators with broad therapeutic potential.

Uncovering mechanisms and predicting tumor cell responses to CAR-NK cytotoxicity is essential for improving therapeutic efficacy. Currently, the complexity of these effector-target interactions and the donor-to-donor variations in NK cell receptor (NKR) repertoire require functional assays to be performed experimentally for each manufactured CAR-NK cell product and target combination. Here, we developed a computational mechanistic multiscale model which considers heterogenous expression of CARs, NKRs, adhesion receptors and their cognate ligands, signal transduction, and NK cell-target cell population kinetics.
