This case report presents a comprehensive evaluation of the complex balance of therapeutic benefits and potential risks associated with the cystic fibrosis transmembrane conductance regulator (CFTR) modulator elexacaftor/tezacaftor/ivacaftor (ETI) therapy in managing an eight-year-old male with cystic fibrosis (CF) and exocrine pancreatic insufficiency (EPI). While ETI therapy significantly enhanced exocrine pancreatic function, it led to hepatotoxicity, necessitating therapy discontinuation. Attempts to restart ETI at reduced doses were unsuccessful due to persistent hepatic dysfunction.

Polyolefins, including high-density polyethylene (HDPE) and isotactic polypropylene (iPP), account for over half of the worldwide plastics market and have wide-ranging applications. Recycling of these materials is hindered due to separation difficulties as co-mingled blends of HDPE and iPP often exhibit brittle mechanical behavior because phase separated domains detach under stress due to low interfacial adhesion. Motivated to improve mechanical properties of mixed recyclates during processing, this work examines the effect of shear on the crystallization kinetics and rheological properties of HDPE-iPP blends utilizing a combination of differential scanning calorimetry (DSC), rheo-Raman spectroscopy, polarized optical microscopy, and scanning electron microscopy (SEM).

Tumor-associated inflammation drives cancer progression and therapy resistance, often linked to the infiltration of monocyte-derived tumor-associated macrophages (TAMs), which are associated with poor prognosis in various cancers. Metastasis causes most cancer-related deaths; however, the efficacy of anti-metastatic drugs is limited by incomplete understanding of the biological mechanisms that drive metastasis. Focusing on the mechanics of metastasis, we propose that the ability of tumour cells to survive the metastatic process is enhanced by mechanical stresses in the primary tumour microenvironment that select for well-adapted cells. In this Perspective, we suggest that biophysical adaptations favourable for metastasis are retained via mechanical memory, such that the extent of memory is influenced by both the magnitude and duration of the mechanical stress.
