Objective: Pedal medial arterial calcification (pMAC) is associated with major amputation in patients with chronic limb-threatening ischemia (CLTI). We hypothesize that this association would be related to unresolved distal ischemia. We investigated relationships across pMAC score, hemodynamic change, and major amputation after infrainguinal revascularization for CLTI.

Article Synopsis
  • - The study compares loop-extrusion and phase-separation as mechanisms for shaping chromosome organization, evaluating their effectiveness individually and together in terms of chromatin structure.
  • - Through the analysis of multiplexed FISH data in human cell lines, both mechanisms demonstrated a strong fit with bulk Hi-C and single-molecule chromatin conformations, especially showing the impact of phase-separation.
  • - The findings suggest that combining loop-extrusion with phase-separation models effectively explains chromatin architecture, highlighting that these mechanisms can work together in single cells while accounting for variability due to epigenetic differences and thermodynamic factors.

Neuroblastoma (NB) is the most common extracranial malignant tumor in children. Although the survival rate of NB has improved over the years, the outcome of NB still remains poor for over 30% of cases. A more accurate risk stratification remains a key point in the study of NB and the availability of novel prognostic biomarkers of "high-risk" at diagnosis could help improving patient stratification and predicting outcome.

Circulating microRNAs (miRNA) have been proposed as specific biomarkers for several diseases. Quantitative Real-Time PCR (RT-qPCR) is the gold standard technique currently used to evaluate miRNAs expression from different sources. In the last few years, digital PCR (dPCR) emerged as a complementary and accurate detection method.

Post-operative atrial fibrillation (POAF), defined as new-onset AF in the immediate period after surgery, is associated with poor adverse cardiovascular events and a higher risk of permanent AF. Mechanisms leading to POAF are not completely understood and epicardial adipose tissue (EAT) inflammation could be a potent trigger. Here, we aim at exploring the link between EAT-secreted interleukin (IL)-1β, atrial remodeling, and POAF in a population of coronary artery disease (CAD) patients.

Article Synopsis
  • The study looked at open abdominal aortic surgery cases from 2007 to 2020, focusing on open aortic aneurysm repairs (OAR), and found that a lot of these surgeries weren't being tracked properly by existing guidelines.
  • Out of 481 surgeries, about 355 were OAR, and more than half of them were complex cases that didn't fit the usual criteria for quality checks.
  • The findings suggest that a new "complex OAR" module should be created to help collect data on these complex surgeries, which could improve the understanding of their outcomes and how many surgeries are being done.

Objectives: There are few contemporary data regarding health-related quality of life (HRQOL) measures in patients with chronic limb-threatening ischemia (CLI).

Methods: The Best Endovascular versus Best Surgical Therapy in Patients with CLI (BEST-CLI) trial is an ongoing, National Institutes of Health-sponsored, multicenter, randomized, controlled trial comparing revascularization strategies in patients with CLI. BEST-CLI baseline HRQOL measures were evaluated for patient-specific variables that were associated with poor HRQOL and then compared with published outcomes.

In recent years, several studies have been published on the prognosis of children with congenital solitary kidney (CSK), with controversial results, and a worldwide consensus on management and follow-up is lacking. In this consensus statement, the Italian Society of Pediatric Nephrology summarizes the current knowledge on CSK and presents recommendations for its management, including diagnostic approach, nutritional and lifestyle habits, and follow-up. We recommend that any antenatal suspicion/diagnosis of CSK be confirmed by neonatal ultrasound (US), avoiding the routine use of further imaging if no other anomalies of kidney/urinary tract are detected.

  • In 2019, new guidelines were created to help understand a condition called chronic limb-threatening ischemia (CLTI) and a concept called limb-based patency (LBP), which means keeping blood flow in a targeted artery after treatment.
  • The study looked at patients from 2016 to 2019 who had surgery for CLTI, focusing on whether keeping LBP was connected to serious problems like amputations or surgeries after treatment.
  • Out of 184 limbs studied, many patients had diabetes and severe tissue loss; results showed that losing LBP was a strong predictor of future serious limb problems.

Visual texture is an important cue to figure-ground organization. While processing of texture differences is a prerequisite for the use of this cue to extract figure-ground organization, these stages are distinct processes. One potential indicator of this distinction is the possibility that texture statistics play a different role in the figure vs.

  • The CBX2 protein, part of the PRC1 complex, is found to be overexpressed in leukemia and may play a significant role in cancer cell survival and leukemic pathogenesis.
  • Researchers used reverse genetic methods and various assays to study the effects of silencing CBX2 in human leukemia cell lines, discovering that reduced levels lead to decreased cell proliferation and increased apoptosis.
  • The study indicates that targeting CBX2 could alter epigenetic regulation and transcriptional programs in leukemia, suggesting that the CBX2-p38 MAPK pathway may be a promising target for new therapies in acute myeloid leukemia (AML).

In Fall 2020, universities saw extensive transmission of SARS-CoV-2 among their populations, threatening health of the university and surrounding communities, and viability of in-person instruction. Here we report a case study at the University of Illinois at Urbana-Champaign, where a multimodal "SHIELD: Target, Test, and Tell" program, with other non-pharmaceutical interventions, was employed to keep classrooms and laboratories open. The program included epidemiological modeling and surveillance, fast/frequent testing using a novel low-cost and scalable saliva-based RT-qPCR assay for SARS-CoV-2 that bypasses RNA extraction, called covidSHIELD, and digital tools for communication and compliance.

Background: Biologics are currently one of the main treatment options for a number of diseases. The IgG4 monoclonal antibody dupilumab targets the Interleukin-4 receptor alpha chain, thus preventing the biological effects of the cytokines IL-4 and IL-13, that are essential for the Th2 response. Several controlled trials showed that dupilumab is effective and safe in patients with atopic dermatitis (AD), severe asthma and chronic rhinosinusitis with nasal polyps (CRSwNP), thus resulting in approval by regulatory agencies.
