Ranitidine disposition has been studied in 12 patients with renal impairment following 50 mg given intravenously and 150 mg given by mouth on separate occasions. The clearance of ranitidine from plasma (y) was correlated with creatinine clearance (x): y = 10.47 + 0.

The expanding pool model of radiocalcium kinetics has been used in 13 maintenance dialysis patients to measure bone mineralization rate. No difficulties were met in applying the data to the model, and values for the bone mineralization rate ranged from 0.0 to 2.

To investigate the possible uptake of aluminium by plasma proteins, the Group IIIB element gallium (67Ga) was used as an analogue. Gel filtration confirmed that plasma bound 67Ga in the transferrin region, and this effect could be reproduced using pure transferrin. Approximately 75%, of 67Ga could be displaced by 10 mumol/l aluminium in the buffer.

An examination of Autographa californica nuclear polyhedrosis virus DNA revealed the presence of five interspersed regions, rich in EcoRI restriction sites, which shared homologous sequences. These homologous regions (hr), designated hr(1) to hr(5), occur at or near the following EcoRI fragment junctions: hr(1)EcoRI-B-EcoRI-I (0.0 map units); hr(2), EcoRI-A-EcoRI-J (19.

Cloned rabbit beta-globin genes, modified in vitro by restructuring or site-directed mutagenesis, were introduced into mouse 3T6 cells, and the resulting transcripts were analyzed by nuclease S1 mapping. The first 109 bp preceding the cap site sufficed for maximal beta-globin transcription. This segment contained three functionally important regions of the ATA box region; the CCAAT box region; and the -100 region.

Cytoplasmic poly(A)(+) RNA isolated from Spodoptera frugiperda cells late after infection with Autographa californica nuclear polyhedrosis virus (30-40 hr pi) was fractionated according to size on denaturing methyl mercury gels. Two major RNA species (1.4 kb and 0.

Organotypic dorsal root ganglion (DRG) cultures were established from all the embryos of two trembler (Tr/+) female mice mated to normal (+/+) males to determine if the trembler mutation would be expressed in nerve tissue culture. Dorsal root ganglia from normal mice maintained in our culture system exhibit substantial myelination after 6 weeks of growth. This normal pattern was observed in approximately one half of the cultures in the present series.

A restriction fragment library containing Autographa californica nuclear polyhedrosis virus (AcNPV) DNA was constructed by using the pBR322 plasmid as a vector. The library, which is representative of more than 95% of the viral genome, consists of 2 of the 7 BamHI fragments, 12 of the 24 HindIII fragments, and 23 of the 24 EcoRI fragments. The cloned fragments were characterized and used to generate physical maps of the genome by hybridizing nick-translated recombinant plasmid to Southern blots of AcNPV DNA digested with SmaI, BamHI, XhoI, PstI, HindIII, and EcoRI restriction endonucleases.

A recombinant cDNA plasmid containing Xenopus immunoglobulin heavy chain sequence has been constructed from Xenopus spleen poly(A)-containing RNA. The plasmid was identified by colony hybridization and a hybridization-translation assay and its identity was confirmed by DNA sequence analysis. The portion of the heavy chain sequence contained in the plasmid is 35% homologous to mammalian mu and gamma sequences.

Aspects of renal bone disease.

In advanced chronic renal failure hypocalcaemia can be due to a combination of factors. These include decreased delivery of calcium into the plasma and increased renal tubular leakage of calcium. The secondary hyperparathyroidism that follows the hypocalcaemia leads to osteosclerosis, probably by movement of cortical mineral into trabecular bone.

Twenty-two virion polypeptides (VP) were detected reproducibly when the occluded form (OV) of [35S]methionine-labeled Ac-NPV, purified from polyhedral inclusion bodies (PIB), was analyzed by polyacrylamide gel electrophoresis and autoradiography. Ten of the VPs and the polyhedral protein (PP) were phosphorylated and 6 VPs were glycoproteins. Purified, nonoccluded virus (NOV) revealed 25 polypeptides.

Total poly(A)-containing mRNA isolated from Xenopus spleens was translated in a rabbit reticulocyte lysate in vitro protein-synthesizing system. Approx. 1% of the radioactivity incorporated into the protein was precipitated by an antibody directed against adult Xenopus IgM.

During a four year period 6 patients from the total of 120 on home dialysis developed pericarditis with effusions. Plasma fibrinolytic activity, measured by euglobulin clot lysis time, was significantly reduced in these patients compared with either normal controls or dialysis patients without pericarditis. These dialysis patients without pericarditis as a group tended to have reduced plasma fibrinolytic activity compared with controls, but the difference was not significant.
