Article Synopsis
  • Sox8 is a transcription factor essential during development and part of the Sox gene family, which is linked to human developmental disorders.
  • Despite previous findings showing minimal effects in Sox8 mutant mice, new analyses indicate that Sox8 deficiency leads to progressive male infertility characterized by irregular spermatogenesis.
  • The absence of SOX8 disrupts Sertoli cell function, causing abnormal placement of germ cells and resulting in sperm with poor motility.

SOX8 is a transcription factor that belongs to the SoxE group of high mobility group (HMG) superfamily of genes. As Sox8 is widely expressed during development, and has male-specific expression at the critical period for sex determination, it was initially hypothesized that its inactivation would result in abnormal sex determination. Though knockout males showed normal sexual development, they did however display a progressive seminiferous tubule failure and infertility.
