The present study was designed to investigate whether hyperhomocysteinemia (HHcy) induced by methionine supplementation promotes oxidative stress and nuclear factor kappaB (NFkappaB) activation in livers of C57BL/6 mice when fed a 2% methionine and low folate (1 mg/kg) diet for 12 weeks. Plasma homocysteine concentrations of mice fed methionine were found to be 49 micromol/L by 12 weeks of feeding, which was five times higher than that of controls. Granulocyte-Macrophage colony stimulating factor (GM-CSF) and Granulocyte colony stimulating factor (G-CSF) are cytokines involved in the differentiation of bone marrow progenitor cells into myeloid cells. A polysaccharide fraction, AIP1, purified from Artemisia iwayomogi was shown to have immunomodulating and anti-tumor activities in mice. In order to determine how the AIP1 fraction exhibits the immunomodulating activity, the effect of the fraction on the apoptosis of mouse spleen cells was investigated. Treatment of the mouse spleen cells with the AIP1 fraction resulted in the suppression of apoptotic death and an extension of cell survival in culture, indicating that the fraction might modulate the death of spleen cells.
