Method: Participants were 313 youth (49% boys; 70.9% European American) and their parents who participated in a longitudinal cohort-sequential study on the development of EC in preschool and its associations with subsequent health outcomes.
Performance of a task involves the engagement of various brain areas, as evidenced by the effects of lesions of particular brain areas and the results of functional neuroimaging and neurophysiological studies. Here we tested the hypothesis that overall task performance would depend on the level of ongoing, resting-state change in synaptic activity of participating areas, such that the degree of success of the outcome would be higher, the higher the resting-state activation. Methods: Kronos Early Estrogen Prevention Study (KEEPS) was a multicenter, randomized, double-blinded, placebo-controlled 4-year mHT trial (oral conjugated equine estrogens or transdermal 17β-estradiol). KEEPS continuation was an observational follow-up of the participants 10 years after the end of mHT.
Medications to treat substance use disorders remain suboptimal or, in the case of stimulants and cannabis, non-existent. Many factors have contributed to this paucity, including the biological complexity of addiction, regulatory challenges, and a historical lack of enthusiasm among pharmaceutical companies to commit resources to this disease space. Despite these headwinds, the recent opioid crisis has highlighted the devastating consequences of SUDs for both individuals and society, stimulating urgent efforts to identify novel treatment approaches.

Resistance to chimaeric antigen receptor (CAR) T cell therapy develops through multiple mechanisms, most notably antigen loss and tumour-induced immune suppression. It has been suggested that T cells expressing multiple CARs may overcome the resistance of tumours and that T cells expressing receptors that switch inhibitory immune-checkpoint signals into costimulatory signals may enhance the activity of the T cells in the tumour microenvironment. However, engineering multiple features into a single T cell product is difficult because of the transgene-packaging constraints of current gene-delivery vectors.
